Objectives Through the observation of chronic pelvic inflammatory disease model rats of behavioral change,macroscopic observation pelvic general situation,morphological changes of uterus tissue in rat were observed under light,detection of serum IL-10,TGF-?1 level,observation of uterus tissue in rat the expression of MMP-2 and TGF-?1 case,discusses on the high's number one party of chronic pelvic inflammatory disease(uterus tissue)in rats,therapeutic effect and mechanism of action.Methods 75 only 12 weeks SD female rats were randomly divided into blank group(10)and module(65),building module adopts 0.06 ml dose chemical burns + 25% phenol gel mechanical damage method of manufacturing the rat model of chronic pelvic inflammatory disease,building 10 days after the end of a random sample of 5 rats pelvic uterine pathological examination,conforms to the pelvic inflammatory disease pathology,suggest building a successful,more than 60 rats were divided into model group,high's number one side high,medium and low dose group,proprietary Chinese medicine Pu Ling basin is phlogistic,particles,western medicine placenta tissue fluid intramuscular injection of oral group,High,medium and low doses of gao shi yifang and pu lingpan yankang granules were given respectively by intragastric administration.The placental tissue fluid intramuscular injection group was given placental tissue fluid intramuscular injection group,the model group was given normal saline intragastric administration,and the blank group was given free water.The behavioral changes of rats,the gross situation of pelvic cavity were observed by naked eyes,the morphological changes of endometrium tissue of rats were observed by light microscope,the levels of serum il-10 and TGF-?1 were detected,and the expressions of mmp-2 and TGF-?1 in rat uterine tissue were detected.Results 1 Behavioral changes of rats: rats in the blank group showed normal physiological functions in all aspects.In the model group,the rats gradually showed mental mania,burnout,dark fur,depilation,less food,more vaginal secretions,loose stools,brown color,and weight loss after modeling.After the end of the model building,manic symptoms began to appear in each group,and there were many vaginal secretions.Gradually,after the drug was given to the stomach,the spirit was slightly improved,hair loss was reduced,vaginal secretions were reduced,and the body weight first decreased and then increased.Among them,the recovery was most obvious in the rats in the high-dose and medium-dose groups of gaos no.1 prescription.2 Morphology of rat uterine tissue observed by naked eyes: in the blank group,uterine morphology was usually large,with rosy color and good elasticity,without deformation,necrosis,hyperemia,edema,etc.,and no adhesion with surrounding connective tissue;In the model group,the uterus showed obvious swelling,severe hyperemia and swelling,pale color,poor elasticity and hardness,intrauterine adhesion and stenosis,and a large amount of effusion was observed in the uterine cavity,which was adhered to the surrounding tissues and partly formed a mass.The uterine morphology was slightly thickened,the elasticity was relatively low,part of the uterus was hyperemic,and there was a little adhesion with the surrounding tissues.The high and medium dose group and the placenta group had similar performance as the blank group.3 Observation of rat uterine tissue morphology under light microscope: blank group:normal uterine wall structure and cell morphology,no obvious hyperemia and edema,occasionally inflammatory cell infiltration.Model group: uterine cavity wall thickening,wall structure disorder,adhesion,cell morphology disorder,visible a large number of inflammatory cell infiltration,epithelial cells degeneration necrosis off,part of the whole layer of fibroblast proliferation.The high and medium dose group and the placenta group had similar performance as the blank group.Low dose of gao shi no.1 prescription and pu ling pan yan kang group: uterine wall was slightly thickened,a small number of inflammatory cells were slightly infiltrated,and the glands were slightly dilated.4Pathological score of uterine tissue: compared with the blank group,the positive score of uterine tissue fat staining in the model group was statistically significant(P<0.05),indicating that the model of chronic pelvic inflammatory disease was successfully established.5 Degree of swelling in rats: the degree of swelling in the model group was significantly higher than that in the normal group(P<0.01)indicating that the model of pelvic inflammatory disease was successfully established.After treatment,the uterine swelling degree of the high,medium and low dose group of gao shi yifang was significantly reduced compared with the model group,and the difference was statistically significant(P<0.05).6 Immunohistochemical method to detect the rat uterus MMP-2,TGF-?1protein expression of uterus tissue in rat model group reduced expression of TGF-beta 1,higher expression of MMP-2,compared with the blank group difference was statistically significant(P<0.05),the basin of gause inflammation side one side high,medium and low dose group,the expression of MMP-2 increases,reduced expression of TGF-beta 1,compared with model group differences were statistically significant(P<0.05).7 Serum levels of TGF-?1 and TGF-?1in rats were detected by ELISA: the serum levels of il-10 and TGF-?1 in rats in the model group were decreased,and the differences were statistically significant(P<0.05)compared with those in the blank group;the levels of il-10 and TGF-?1 in the high,medium and low dose groups were increased,and the differences were statistically significant(P<0.05)compared with those in the model group.8 In vitro bacteriostasis test results: gao shi ban yan fang yifang has different degree of inhibition on staphylococcus aureus,hemolytic streptococcus b and escherichia coli.Conclusions 1 The model was established by chemical burns with a dose of 0.06 ml of25% phenol gel plus mechanical injury,and the behavioral changes,uterine and ovary mass and body weight of the rats were all changed.The wall of uterine cavity was thickened,the wall structure was disorganized,the adhesion was disorganized,and the cell morphology was disorganized.The expression of inflammatory cytokines was abnormal and the rat model of chronic pelvic inflammation was successfully prepared.2 gao shi no.1 can effectively treat chronic pelvic inflammatory disease,can play a better antiinflammatory,repair damaged tissue;Among them,the high and medium dose group of gaoshipiyan prescription no.1 was equivalent,superior to the low dose group of gaoshiyan prescription no.1,and similar to the effect of the placental tissue fluid group.The lowdose group of the first prescription of gaoshi penyan prescription had the same effect as the puling penyan kang granule group.3 no.1 of gao shi can regulate the pre-inflammatory cytokines in rats with chronic pelvic inflammation,regulate the expression of various inflammatory factors in tissues and blood,and make it tend to be normal,so as to play a therapeutic role in alleviating and eliminating inflammation and inhibiting pelvic adhesion.In addition high shi yifang can be opposite aureus staphylococcus,hemolytic streptococcus b,escherichia coli all have the inhibition ability of different level,have the effect that fight phlogistic eliminate bacterium.Figure 22;Table 7;Article 83 refs.
