Objective: In order to provide the theory basis for the expansion and utilization ofthe medicinal resource of Vitex trifolia L. var. simplicifolia Cham., the chemicalconstituents of the leaf was analyzed here. The quality control methods of Fructus viticiswere also studied.Methods:70%ethanol reflux extraction was repeated3times,5hrs each andethanol was recovered. Suspension was extracted with petroleum ether, ethyl acetate,n-butanol respectively. the ethyl acetate fraction was separated and purified by columnchromatography and preparative liquid equal separation technique. Purified compoundswere identified with1H-NMR,13C-NMR and other spectroscopic techniques todetermine its structure. On the other hand, in order to shape a quality control system andexplore appropriate method for quality control of Fructus viticis, Vitexicarpin level wasselected for reference to evaluate influence of different factor via determination byHPLC.Results:15compounds were separated by IR, UV, ESI-MS,1H-NMR,13C-NMRand other spectroscopic techniques and8chemical structures were identified by physical,chemical methods. They were Vitexicarpin(II), Luteolin(III), Luteolin-7-O-glucoside(V),ρ-coumaric acid(VI),3,6,7-trimethylquercetagetin (VII),4-Hydroxybenzoic acid (IX),3,4-Dihydroxybenzoic acid (X), Methyl transphydroxycinnamat(XII). In Viticis flavin asan index were measured and found the fruits of Vitex trifolia L. var. simplicifolia Cham atthe end September were assessed to have highest content of Viticis flavin; In rawFructus viticis Vitexicarpin content was significantly higher than Pharmacopoeia standard of0.030%, and that in fried product it’s below0.030%;Wild Vitex moved fromthe barren sea or lake sand to a much more fertile experimental field to acclimate for1year, the average content Vitexicarpin in fruit decreased by56.1%; Comparing to thecontrol, Vitexicarpin content increased34.76%in friut of28d water stress treatmentbefore harvest, and Vitexicarpinin7d,14d,21d treatment it was significantly lower than28d.Conclusion: The chemical composition were similar between fruits and leaves ofVitex trifolia. The suitable time to harvest the Vitex trifolia fruit is from the end ofAugust to early October in Shandong and surrounding provinces. The Vitexicarpincontent decreased after fried, so it ought to be used in raw. Different habitat acclimationof12months can significantly reduce Vitexicarpin level in friut. Comparing to thecontrol group, long time drought stress could facilitate accumulation of Vitexicarpinbefore harvest while significantly decrease the growth of new shoots.
