Objective:To investigate the polymorphism,the distribution and the allelomorph frequency of the IL-33 genetic locus rs4740840 and rs16924159,and to analyze the relationship between IL-33 serum level and asthma in Heiyi Zhuang pediatric population.Methods:This was a case–control study of 118 asthmatic children and 121 healthy controls.The polymorphisms of rs4740840G/A and rs16924159G/A of IL-33 gene were analyzed with imLDR multiplex SNP,and the serum level of IL-33 was detected by ELISA.SPSS 24.0 was applied in statistical analysis.Direct counting method was used in the distribution frequency of genotype and allele in the asthma group and the healthy control group.Genotypic and allelic frequencies between patients and controls was compared by?~2test.The risk assessment of asthma was expressed by odds ratio(OR)and 95%CI.Serum IL-33 levels among genotypes were compared by?x±s.Independent samples T test was uesd to test the comparison between groups.One way analysis of variance(One way ANOV)for intra-group comparison.Statistical significance was assumed at the P<0.05 level.Hardy–Weinberg equilibrium was used for Group representation.Results:(1)The average age was 7.3±2.4 in children with asthma[68(57.6%)males and 50(42.4%)females]and 7.6±2.5 in controls[76(62.8%)males and 45(37.2%)females].There was no statistical difference between children with asthma and controls concerning age(t=-0.779,P=0.437)and gender(?~2=0.67,P=0.413).(2)There were three genotypes in the rs4740840 of the IL-33 gene:AA,GA,GG,the genotype frequencies were 35.6%,40.7%,23.7%in the asthma group and 21.5%,46.3%,32.2%in the control group.There was significant difference in the distribution of genotypes between the two groups(P<0.05).the risk of asthma in children with AA genotype was significantly higher than that of GG genotype(P<0.05).The allele frequencies of A were 55.9%(G 44.1%)in the asthma group.A44.6%.The allele frequencies of A were 55.4%(G 44.6%)in the control group.There was significant difference in the distribution of allelic frequencies between the two groups(P<0.05).(3)There were three genotypes in the rs16924159 of the IL-33 gene:AA,GA,GG,the genotype frequencies were 10.2%,35.6%,54.2%in the asthma group and 5.8%,34.7%,59.5%in the control group.There was no significant difference in the distribution of genotypes between the two groups(P>0.05).The allele frequencies were 28.0%(A)and72.0%(G)in the asthma group and 23.1%(A),76.9%(G)in the control group.There was no significant difference in the distribution of allelic frequencies between the two groups(P>0.05).(4)The ELISA results showed that the serum levels of interleukin-33 in the asthma group were significantly higher than those in the control group(P<0.05).The results of the tests of the genotype AA,GA,GG of rs4740840G/A single nucleotide polymorphism showed that there was significant difference among the three genotypes and the serum levels of IL-33 in case group(P<0.05),and the level of serum IL-33 in AA genotype was higher than that of GG and GA genotypes.The serum IL-33 levels of AA and GA genotypes in asthma group were higher than those in healthy control group(P<0.05).Conclusion:The polymorphisms is different in IL-33 gene of rs4740840 and rs16924159 in race distribution,and the genotypes and alleles of IL-33 gene are also different in race distribution.The rs4740840G/A polymorphism of IL-33 gene is associated with the susceptibility of asthma in Heiyi Zhuang children.Nevertheless,rs16924159G/A polymorphism may have a connection with asthma.The rs4740840G/A polymorphism of IL-33 gene and the serum level of IL-33 were associated with asthma in Heiyi Zhuang children in Guangxi.
