A Study On The Correlation Between The Gene Polymorphism And Serological Level Of IL-6R And Asthma In Children With Heiyi Zhuang In Guangxi

Objective:To investigate the relationship between the locus polymorphisms of rs2228145,rs12083537,rs4845617,rs4845374,rs2229238 of IL-6R gene and the development of asthma in children with Heiyi Zhuang in Guangxi.The distribution of the above five loci in children and other ethnic groups of polymorphism in Guangxi and the relationship between these five loci and the susceptibility to asthma in children of Heiyi Zhuang in Guangxi were analyzed.To analyze the serum characteristics of s IL-6R in children with asthma in Heiyi Zhuang in Guangxi.Methods:The polymorphisms of rs2228145,rs12083537,rs4845617,rs4845374 and rs2229238 in 236 subjects(121 Heiyi Zhuang children with asthma in Guangxi and 115 healthy Heiyi Zhuang children in Guangxi)were genotyped by SNa Pshot technique.Serum levels of s IL-6R were detected by enzyme linked immunosorbent assay(ELISA).The data were analyzed with SPSS 23.0 statistical software.The genotype frequency and allele frequency of the above 5 loci were measured by direct counting method in Guangxi Heiyi Zhuang asthma group and healthy group.Hardy-Weinberg Equilibrium(HWE)law was used to test the population representativeness.?2 genotype distribution and allele distribution of Heiyi Zhuang asthma group and healthy group were examined and compared.The risk of asthma was expressed as a ratio(OR)and a 95% confidence interval(CI).Serum s IL-6R levels were measured by median and interquartile intervals,and the rank sum test was performed by two independent samples in asthma and health groups.The statistical analysis showed that P<0.05 had statistical significance.Results:(1)Among the 121 children with asthma,75(62.0%)were males and 46(38.0%)were females.The ratio of males to females was 1.63:1.The age range was 0.5-12 years with an average age of 4.1±2.9 years.There were 115 healthy Heiyi Zhuang children: 64(55.7%)males,51(44.3%)females,the ratio of males to females was 1.25:1.Age 0.5-13 years,mean age 4.0 ± 3.6 years.There was no significant difference in gender and age composition between the two groups(P>0.05).(2)Three different genotypes were detected at each of the five loci.Three genotypes of AA / AC / CC were detected at rs2228145 locus in Guangxi Heiyi Zhuang asthma group and healthy group.AA / AG / GG genotypes were detected at rs12083537,rs4845617 loci in Guangxi Heiyi Zhuang asthma group and healthy group.Two genotypes(TT / AT)were detected at rs4845374 locus in Guangxi Heiyi Zhuang asthma group and three genotypes(TT / AT / AA)were detected in Guangxi Heiyi Zhuang Health Group.Three genotypes of CC / CT / TT were detected at rs2229238 locus in Guangxi Heiyi Zhuang asthma group and healthy group.(3)The allele frequency distribution of some of the above five loci showed racial differences.(4)The genotype frequencies of rs2228145 of IL-6R gene in asthma group were AA 66.9%,AC 31.4% and CC 1.7%,and the allele frequencies were A 82.6% and C 17.4%,respectively.In Guangxi Heiyi Zhuang healthy group,the genotype frequencies were AA 52.2%,AC 40.9% and CC 6.9%,and the allele frequencies were A 72.6% and C 27.4% respectively.There were significant differences in genotype and allele frequency(P<0.05).Compared with CC genotype,the risk of asthma in AA genotype was significantly higher(?2=5.290,P=0.021;OR=5.4,95%CI=1.107-26.349),the risk of asthma of A allele was 1.796 times higher than that of the C allele(OR=1.796,95%CI=1.156-2.793).(5)There was no significant difference in genotype and allele frequency of rs12083537,rs4845617,rs4845374,rs2229238 of IL-6R gene between Guangxi Heiyi Zhuang asthma group and health group(P>0.05).(6)The level of serum s IL-6R in Guangxi Heiyi Zhuang asthma group was higher than that in healthy group,the difference was statistically significant(P<0.05).Conclusion:(1)rs2228145,rs12083537,rs4845617,rs4845374,rs2229238 are the polymorphisms of IL-6R gene.The allele frequencies of rs2228145,rs12083537 and rs4845374 were different among different races.(2)rs2228145 polymorphism of IL-6R gene is associated with asthma susceptibility in Guangxi Heiyi Zhuang children,AA genotype is susceptible genotype,while rs12083537,rs4845617,rs4845374,rs2229238 may not be associated with asthma in Guangxi Heiyi Zhuang children.(3)The increase of s IL-6R level may be related to the occurrence of asthma in Guangxi Heiyi Zhuang children.
