Objective: To detect GPC1 expression and distribution profile in liver tissues of biliary atresia(BA)infants;To fulfill genetic etiology of BA,and To provide theoretical basis for early diagnosis of BA.Methods: 35 infants(male/female,14/21)with BA and 42 infants(male/female,14/25)with choledochal cyst(CC)that were admitted and treated in Shenzhen Children's Hospital between July 2016 and February 2018 were enrolled in the study as an experiment group and a control group,respectively.All the liver samples collected were divided into three pieces.One was put in formalin,while the other two were put in RNALater and stored in-80?.Three procedures were performed on the above liver tissues.1.IHC was performed on formalin-stored liver tissues from experiment group to identify distribution of GPC1 in BA livers.2.Total RNA was retracted from RNA-stored liver tissues of both groups and subsequently qPCR were performed to clarify whether there was any difference on the expression profile of GPC1 at mRNA level.3.Total protein was retracted from RNA-stored liver tissues of both groups,followed by applying Western Blot(WB)to test any difference on the expression characteristics of GPC1 at protein level.Results: 1.GPC1 located at cholangiocytes in BA liver tissues.2.The expressionsof GPC1 at mRNA level decreased in BA liver tissues compared to CC liver tissues(0.65±0.05 vs.2.542±0.12,p?0.05).3.There was a significant difference on the expression profile of GPC1 at protein level in BA relative to CC(1.31±0.14 vs.2.83±0.25,respectively).Conciusion: GPC1 existed at bile duct cells,and it was down-expressed in BA livers.Its expression profile is closely related to the development of BA,which provides a new clue for the genetic theory of the etiology of biliary atresia.
