Objective:To investigate the expression of PAK6(p21-activated kinase 6)protein and its significance in urinary bladder cancer tissue.Methods:Immunohistochemistry was used to analyze the positive expression of PKA6protein in bladder cancer tissue from 89 patients with bladder cancer(cancer group)and the normal epithelium tissue from 30 patients undergoing total or partial cystectomy(control group).The expressions of PAK6 mRNA and protein were detected by Real-time fluorescence quantitative PCR and Western blotting respectively in Biu-87cells、T24 cells and SV-HUC-1 cells.Result:The positive expression of PKA6 protein was significantly lower in bladder cancer group(46.07%)than that in control group(96.67%)(P<0.05),and in patients with recurrence(29.41%)than that in patients with primary cancer(56.36%)(χ~2=6.143,P=0.013),while no significant difference were found in the number,pathological grade,metastasis or not and the clinical stage of bladder cancer(P>0.05).The expression levels of PAK6mRNA and protein were reduced gradually in Biu-87cells(0.0189±0.0037、0.0530±0.0129)、T24 cells(0.0993±0.0121、0.1431±0.0081)and SV-HUC-1 cells(1.0403±0.3755、0.3381±0.0511)(P<0.05).Conclusion:PAK6 protein is lowly expressed in bladder cancer and is closely correlative to recurrence of bladder cancer,playing a inhibitory role in the development and progress of urinary bladder cancer.Therefore it could be used as a antitumor target.
