Swertiae Herba called Zihua Dangyao consists of Swertia pseudochinensis Hara which is reaped in summer and autumn.Swertiae Herba called“Ganyancao”is used as a traditional herb in Hebei and Neimenggu Provinces and is recorded in Ch.P?1977?,Ch.P?2010?and Ch.P?2015?.It is tasted bitter and cold and considered belong to liver,stomach and large intestine meridian.It can be used in damp-heat jaundice,hypochondriac pain dysentery and inappetence.The main component of Dangfeiligan Capsule is Swertiae Herba.It is recorded the resource,character,processed method,flavor,meridian tropism,indication,identification,examination and the determination of sweroside with Swertiamarin in compendia.With the increasing requiement of Swertiae Herba in related product,the method of pharmacopeia is needed to be proved.The quick and convenient TLC method of identification and HPLC characteristic chromatogram were developed to evaluate the quality of the herb.A total of 20 components were identified in Swertiae Herba including 6 iridoids,3 ketones,8 xanthonoids and flavonoids,an anthraquinone,a triterpenoids and an alkaloid.Loganic Acid and3-O-deMethylswertipunicoside were reported first time in Swertiae Herba.The study had great value to the quality control and chemical study of Swertiae Herba.Part one Research on TLC quality control method of Swertiae HerbaObjective:To establish the quality control method of Swertiae Herba by TLC.Method:The easy,effective and quick method is established with reference herb and reference substance.The thin layer run by upon of petroleum ether?60?90??-ethyl formate-formic acid?15:7:1?solution.Then run 8cm away and dry the board.Spray 10%vitriol in alcohol and color the board with the heat.The pots can be seen in the sunsite.Result:The sample can be greatly recognized in this situation.The TLC method is great for the control of Swertiae Herba.Part two Research on the Characteristic Chromatogram of Swertiae Herba by HPLCObjective:To establish the HPLC Characteristic chromatogram analysis of Swertiae Herba by HPLCMethod:The Waters 2695-2996,Dionex Ultimate 3000 and Agilent1260 high performance liquid chromatograph instrument equipped with Shiseido CAPCELL PAK C18 MG?,Boston Green ODS and Phenomenex Gemini column were used.Methanol-Acetonitrile-water containing 0.05mol·L-1 sodium dihydrogen phosphate?adjust pH value to 4.0 with 0.1%phosphoric acid solution??15:5:80?were used as mobile phase,monitored with the wavelength of 246 nm.Flow rate was set at 1.0 mL·min-1.The main component relative retention time comparison method were used to evaluate characteristic Fingerprints of 10 batches of Swertiae Herba produced in different regions.Result:The HPLC Characteristic chromatogram was completed with three recognizable peaks.The RSD of relative retention times of the peaks from 10 batches of Swertiae Herba less than 0.3%.Part three The chemical analyst of Swertiae Herba by UPLC-Q-TOF-MS/MSObjective:The chemical analyst of Swertiae Herba by UPLC-Q-TOF-MS/MSMethod:Analyst the chemical components of the Swertia pseudo-chinensis Hara by PeakView 2.0,Chemspider and Chemicalbook.Confirm the structures by references and the MS/MS chromatograms.Result:A total of 20 components were identified in Swertiae Herba including 6 iridoids,3 ketones,8 xanthonoids and flavonoids,an anthraquin-one,a triterpenoids and an alkaloid.Loganic Acid and 3-O-deMethylswertip-unicoside were reported first time in Swertiae Herba.
