Objective:Based on study of flavonoids chemical compositons in the root of Sophora alopecuroides L.,analyze targets and related signal pathways about depression based on the ideas of the network pharmacology model,illuminate preliminarily substance basis and molecular mechanism about antidepressant effect of flavonoids chemical compositons in the root of Sophora alopecuroides L.using pharmacological experiments.Methods:1.Modern chromatographic techniques were adopted to separate and enrich flavonoids in the root of Sophora alopecuroides L.2.Based on the network pharmacology,we put forward the possible pharmacological effects and mechanisms of flavonoids chemical compositons in the root of Sophora alopecuroides L.,which comes mainly from documents.3.Antidepressant effect of total flavonoids and Trifolirhizin in the root of Sophora alopecuroides L.was observed by measuring changes of behaviors using CUMS model.4.Based on molecular biology techniques,activities study on Trifolirhizin and total flavonoids to depression related targets and signal pathways will be done.Results:1.Flavonoids chemical compositons isolated from in the root of Sophora alopecuroides L.were Exiguachromone A,Trifolirhizin,Sophoraflavanone G.2.Ingredient-target-pathway network graphs of 11 monomer compositions were showedin the paper,and based on the construction of network pharmacology,flavonoid monomer and total flavonoids in the root of Sophora alopecuroides L.had potential to affect insulin signaling pathway and depression signaling pathway.3.Total flavonoids in the the root of Sophora alopecuroides L.could significant activity of antidepression,obviously reduced immobility time in TST and FST induced by CUMS,and increased protein expression of P-PI3 K,P-Akt,P-mTOR ? P-GSK3?.4.Trifolirhizin could significant activity of antidepression,remarkably reduced immobility time in TST and FST induced by CUMS,increased expression of P-PI3 K,P-Akt,P-mTOR ? P-GSK3?.Conclusion:1.11 flavonoid monomer compositions in the root of Sophora alopecuroides L.had potential to affect insulin signaling pathway,inflammatory signaling pathway and depression signaling pathway.2.It was preliminarily illuminated that total flavonoids in the root Sophora alopecuroides L.exerted antidepressant effect by PI3K/Akt signaling pathway.3.It was preliminarily revealed that Trifolirhizin alleviated depression-like symptoms via PI3K/Akt signaling pathway.
