Analysis On Prognosis Of Patients With EC After Operation And Characterization Of MUC1 Gene In ESCC | | Posted on:2018-07-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y M Wang | Full Text:PDF | | GTID:2404330596989856 | Subject:Chest science | | Abstract/Summary: | PDF Full Text Request | | Objective: 1.We aimed to investigate prognostic factors that affect overall survival in octogenarian patients with esophageal cancer who underwent esophagectomy.2.We used large-scale databases to analyze characterization of MUC1 expression and offered theory basis to clarify the role of MUC1 in ESCC.3.We also analyzed role of MUC1 in cancer chemotherapy resistance.Methods: 1.Retrospective review of 86 patients undergoing esophagectomy foresophagealcancer during Oct.2004-Oct.2014.All patients wereover 80 yearsold.We analyzed the effects of age,sex,complications,tumor T stage,lymph node metastasison overall survival.2.We used ONCOMINE and COSMIC databases to analyze MUC1 gene characteristics.3.We used ONCOMINE database and COSMIC database to analyze MUC1 gene copy numbers and mutations in ESCC,then we used ESCC samples to analyze MUC1 gene mutations,protein expression and MUC1 glycosylation.3.We analyzed function of MUC1 in chemotherapy resistance and relationship between MUC1 and EGFR by Western blot,Realtime-qPCR and CCK8.Results: 1.T stage,longest diameter of tumor,postoperative complications and lymph node metastasis were indispensable risk factors.Patients of T1 and T2 stages had longer survival duration than those of T3 stage.2.MUC1 has high expression levels and few gene mutations in ESCC from ONCOMINE and COSMIC databases.MUC1 O-glycosylation expression levels are higher in tumor tissues than para-carcinoma tissues in 14 ESCC samples.C1GALT1 expression levels are higher in tumor tissues than para-carcinoma tissues in 7 ESCC samples.3.MUC1 mRNA and protein levels can be upregulated by Etoposide and Doxorubicin in HeLa 229 and NCI-H292 cell lines.MUC1 is associated with chemotherapy resistance.These phenomenon can be blocked by EGFR inhibitor Erlotinib.Conclusions: 1.Elderly esophageal cancer patients(>80 years old)with earlier T stage,smaller tumor diameters,less postoperative complications and negative lymph node metastasis can benefit from esophagectomy.2.There are some mutations of MUC1 gene in ESCC samples but they have no effects on MUC1 protein.Glycosylation levels of MUC1 protein are higher in ESCC tumor tissues than para-carcinoma tissues and this phenomenon is associated with C1GALT1.3.MUC1 can enhance chemotherapy resistance in HeLa229 and NCI-H292 cell lines.EGFR inhibitor Erlotinib can block chemotherapy resistance induced by MUC1. | | Keywords/Search Tags: | Esophageal cancer, Esophageal surgery, Prognosis, MUC1, Gene mutation, Glycosylation | PDF Full Text Request | Related items |
