Objective: On the basis of the electro-acupuncture treatment of acute facial nerve injury,the New Zealand rabbits with acute facial nerve injury were studied.The JAK-STAT regulatory pathway closely related to facial nerve injury repair was used to study the different electrical stimulation doses of electro-acupuncture.The influence of JAK-STAT signaling pathway-related molecular expression on facial nerve injury provides a basis for the biologic mechanism of different types of electro-acupuncture to promote the repair of acute facial nerve injury,and provides an experimental basis for the correct selection of electro-acupuncture stimulation.Method: Sixty New Zealand rabbits were randomly divided into blank group,sham operation group,model group,electro-acupuncture light stimulation group,electro-acupuncture medium stimulation group and electro-acupuncture strong stimulation group.Except for the blank group and the sham operation group,the other groups used the facial nerve compression method to replicate the rabbit model of facial nerve injury.The electro-acupuncture treatment group selected “he gu”,“di cang”,“jia che”,“quan liao”,“yi feng”.Five points,acupuncture combined with electro-acupuncture treatment,after treatment,each group of New Zealand rabbits used Histomorphology,ELISA,Western Blot and other techniques to observe the repair effect of electro-acupuncture on acute facial nerve injury under different intensity,frequency and treatment time.The facial nerve tissue morphology,JAK-STAT regulatory pathway signal transduction factor JAK1,STAT3,negative feedback regulators SOCS1,SOCS3 were used as indicator to analyze the different effects of different stimulating doses of electro-acupuncture on acute facial nerve injury repair.Results:1 Facial nerve injury in New Zealand rabbitsIn the blank group,the facial muscles of New Zealand rabbits were normal.After the tentacles,the beards were immediately erected,the tip of the nose was centered,and the mouth was bilaterally symmetrical.In the sham operation group,the facial muscles of New Zealand rabbits were nearly normal.After the tentacles,the beards were immediately erected,the tip of the nose was centered,and the mouth was bilateral.model group New Zealand rabbit facial muscles are slow,rabbit whiskers are not erect,nose tip is opposite to the contralateral side,one side of the mouth is drooping;electro-acupuncture light stimulation group compared with the model group New Zealand rabbit muscle movement,rabbit tentacles after the beard erect,nose tip Centered,the two sides of the mouth were slightly asymmetrical;the electro-acupuncture in the stimulation group and the electro-acupuncture strong stimulation group showed that the facial muscles were slow,the rabbits had no whiskers after the tentacles,the nose tip was opposite to the opposite side,and one side of the mouth was drooping.2 Facial nerve damage morphological changes of facial nerve in New Zealand rabbitsUnder the blank group,the intact facial nerve fiber bundle can be seen,and the bundle membrane is continuous and smooth.Under the sham operation group,the nerve fibers can be seen with different degrees of edema.Under the facial nerve microscope,the nerve fibers can be seen with different degrees of edema and nerve bundles.The integrity of the membrane was destroyed;the intact facial nerve fiber bundle was seen under the light stimulation group,the bundle membrane was continuous and smooth,the vacuolization of the myelin sheath was visible,the staining was uneven,and occasionally there was inflammatory cell infiltration;the middle stimulation group was visible under the facial nerve microscope.Complete facial nerve fiber bundles,nerve fibers can be seen with varying degrees of edema,the tunica albuginea is continuous and smooth,the myelin sheath can be seen as vacuolated,the staining is uneven,and some of them can be demyelinated;in the strong stimulation group,the complete facial nerve fiber bundle can be seen under facial nerve microscopy.The bundle membrane is continuous and smooth,and the myelin sheath is vacuolized,and the staining is uneven,and demyelination can be seen.3 JAK-STAT pathway transduction factor JAK1,STAT3 expression(1)JAK1 expression level of rabbits in each group was comparedCompared with the blank group,the expression level of JAK1 in the model group increased,and the difference was statistically significant(P < 0.05);Compared with the model group,JAK1 expression level in the light stimulation group decreased,with significant difference(P < 0.05);there was significant difference between the medium stimulation group and the light stimulation group(P < 0.05);there was significant difference between the strong stimulation group and the light stimulation group(P < 0.05).(2)The STAT3 expression level of New Zealand rabbits was comparedCompared with the sham operation group,the expression level of STAT3 in the model group was significantly increased(P<0.05).Compared with the model group,the expression level of STAT3 in the light stimulation group was significantly lower,and the difference was statistically significant(P< 0.05);Compared with the light stimulation group,the expression level of STAT3 in the middle stimulation group was significantly different(P<0.05).Compared with the light stimulation group,the expression level of STAT3 was significantly changed in the strong stimulation group(P < 0.05).4 JAK-STAT pathway negative feedback regulators SOCS1,SOCS3 expression(1)Results of SOCS1 statistical analysisCompared with the blank group,there was no significant difference in the expression of SOCS1 protein in the sham operation group(P>0.05).Compared with the sham operation group,the expression level of SOCS1 protein in the model group was significantly increased(P<0.05).Compared with the model group,the expression level of SOCS1 protein in the electro-acupuncture mild stimulation group was significantly lower(P<0.05).Compared with the electro-acupuncture light stimulation group,the expression level of SOCS1 protein in the electro-acupuncture group was significantly higher.The difference was statistically significant(P<0.05).Compared with the electro-acupuncture mild stimulation group,the expression level of SOCS1 protein in the electro-acupuncture strong stimulation group was significantly increased(P<0.05).(2)Results of SOCS3 statistical analysisCompared with the blank group,there was no significant difference in the expression level of SOCS3 protein in the sham operation group(P>0.05).Compared with the sham operation group,the expression level of SOCS3 protein in the model group was significantly increased,and the difference was statistically significant(P< 0.05);the expression level of SOCS3 protein in the electro-acupuncture mild stimulation group was significantly lower than that in the model group(P<0.05).The expression level of SOCS3 protein in the electro-acupuncture light stimulation group was significantly lower than that in the electro-acupuncture stimulation group.The difference was statistically significant(P<0.05).The expression level of SOCS3 protein in the electro-acupuncture mild stimulation group was significantly lower than that in the electro-acupuncture strong stimulation group(P<0.05).Conclusion:(1)Acute facial nerve injury model New Zealand rabbit facial expression muscle dyskinesia,facial nerve tissue morphological damage is obvious,electro-acupuncture light stimulation can significantly improve the facial morphology and facial muscle movement of New Zealand rabbits with acute facial nerve injury model,while electro-acupuncture medium and severe stimulation The change in the above indicators is not obvious.(2)Acute facial nerve injury model The expression of facial nerve JAK1,STAT3,SOCS1 and SOCS3 in New Zealand rabbits is abnormal,indicating abnormal changes of JAK-STAT regulatory pathway in New Zealand rabbits with acute facial nerve injury model;electro-acupuncture light stimulation can significantly regulate JAK-STAT signaling pathway in facial nerve tissue Abnormal expression of these factors indicates that the mechanism of electro-acupuncture light stimulation to promote the repair of acute facial nerve injury is closely related to the regulation of JAK-STAT signaling pathway cytokine expression and signal transduction;in this study,electro-acupuncture medium and severe stimulation of JAK-STAT signal The expression of pathway cytokines is not obvious.
