The species diversity and ecosystem diversity of fungi lead to the diversity of their metabolites.Fungal metabolites become an important source of new drug development and lead drug discovery.With the development of separation technology,more and more fungal secondary metabolites have been discovered.However,many known compounds have been found repeatedly.It is becoming increasingly difficult to find new compounds from fungi.Special eco-environment,caught our attention,due to the novel structures and interesting bioactivities of their secondary metabolite.In this paper,secondary metabolites from two unique ecological niches fungi,mine soil fungus Tolypocladium sp.XL-115 and Preussia isomera XL-1326,an endophytic fungus from panax notoensis,were studied.Thin layer chromatography,silica gel column chromatography,ODS column chromatography,glucan LH-20 gel column chromatography,semi-preparative HPLC,recrystallization and other methods were used for the separation of fungal secondary metabolites.The structure of the compound was analyzed by nuclear magnetism(1H-NMR,13C-NMR,1H-1H COSY,HSQC,HMBC,NOESY),high resolution mass spectrometry,optical rotation,UV,IR,circular dichroism,single crystal X-ray diffraction and other modern analytical techniques.Tolypocladium sp.XL-115 was isolated from the mine soil collected from Lianyuan,Hunan Province,P.R.China.Using OSMAC?One strain many compounds?strategy,we got twelve new prenylated indole diterpene alkaloids,tolypocladins A-L?1-12?,together with a known analogue terpendole L?13?.Compounds 1-10 were tested for antibacterial,antifungal and antitumor activities,and compounds 11-13 and terpendole L were tested for antibacterial,antifungal.Test result suggested that,compounds 1-10,13 showed different degrees and ranges of antifungal activity;compound 1,8,11,13 exhibited different degrees of antibacterial activity;compound1-10 showed antitumor activity.Preussia isomera XL-1326 was isolated from the stems of Panax notoginseng collected from Wenshan,Yunnan Province,People's Republic of China in August 2018.We cultivated XL-1326 in MEB rice solid medium,and got two new compound?+?-preuisolactone A?14a?and?-?-preuisolactone A?14b?,together with seven new prenylated compounds 15-21.?±?-preuisolactone A was tested for antibacterial,antifungal and antitumor activities.It showed antifungal activity against Alternaria alternate and antibacterial activity against Micrococcus luteus and Bacillus megaterium.
