An Animal Experiment Of Intervention By "Recipe To Harmonize And Supple Liver" In Rat Model Of Liver Fibrosis Induced By Carbon Tetrachloride

Objective:To replicate rat model of liver fibrosis induced by carbon tetrachloride?CCl₄?,and to observe the anti-fibrosis effect of“recipe to harmonize and supple liver”?H&SL?on the model rats.Methods:70 rats were subcutaneously injected CCl₄ solution?CCl₄:olive oil=3:2,3ml/kg,2 times/week?into the neck-back as the model group randomly and the other 10 rats without special treatment as the control group or the blank control group?Ctrl?.After successful modeling,the model group was randomly divided into the normal saline group?NS,0.9%sodium chloride solution?,the colchicine group?COL,0.2mg/kg colchicine saline solution?and the high?H-H,19.8g/kg H&RL?,middle?H-M,9.9g/kg H&RL?and low dose groups of H&RL?H-L,4.95g/kg H&RL?.The corresponding test drugs were given by gavage for 4 weeks?1 time/day?.After the course of treatment,body weight changes and liver index were calculated,liver function and coagulation function of serum were measured,fibrosis degree of liver tissue was observed by HE staining,and differences between groups were compared.Results:?1?At the 12th week of modeling,the rats in the model group with a cumulative mortality rate of 20%were smaller in size,listless in spirit,slow in reaction and their stools were thin.The level of serum ALT,AST,TBil,and Alb was found significant abnormal?P<0.001?.The liver pathological images of rats randomed in the model group showed significant fibrosis?F2?.After 4 weeks of placebo intervention,the degree of liver fibrosis did not reverse spontaneously.?2?After 4 weeks of intervention,the general condition of the rats in each intervention treatment group were improved in varying degrees compared with the previous one.The level of serum ALT,AST,and TBil of rats in H-H were basically recovered as normal?P>0.3?,and the serum ALT levels in the other intervention treatment groups were still abnormally high in varying degrees?P<0.05?.Compare to H-L,H-H had lower spleen coefficient,higher plasma PT and INR?P<0.05?,and higher liver weight?P=0.032?.The order of relative improvement degree of liver function in rats was as follows:H-H>H-M>COL>NS>H-L.?3?After 4 weeks of intervention,the order of the relative degree of liver fibrosis in METAVIR score of pathological images of rats in each group was as follows:NS>COL=HL>HM>HH>Ctrl?P<0.05?.Conclusion:The rat model of liver fibrosis can be replicated by subcutaneous injection of CCl₄ solution?CCl₄:olive oil=3:2?into the neck-back,3ml/kg,2 times/week for 12 consecutive weeks,which is suitable for anti-fibrosis experiments in vivo within 4 weeks.To the above model rats,H&SL had significant effects on improving liver function and degree of liver fibrosis in a dose-dependent manner.
