Literature Research On The Treatment Of Alzheimer Disease With Traditional Chinese Medicine

Objective:In this study,we summarize the medication rules by retrieving,collecting and organizing the clinical study literature and animal experiment literature for the treatment of Alzheimer disease(AD)with traditional Chinese medicine and the statistical analysis of traditional Chinese medicine and related classical formulas contained.It can provide data supports for clinical medication and drug research.Method:The literature published from January 1979 to July 2019 was retrieved in China Science and Technology Journal Database,Wanfang Data Knowledge Service Platform,China National Knowledge Infrastructure,Pubmed,EBSCO,Science Direct and so on.We used the CNKI-E Study to remove reduplication and determine the final inclusion of literature according to the literature screening criteria.After the standardization of traditional Chinese medicine names by "greater dictionary of Chinese medicine" and"traditional Chinese pharmacy",the frequency statistics of core drugs and high-frequency classical formulas were conducted using Microsoft Excel 2016,and the screening of core drugs and high-frequency classical formulas was conducted.SPSS 22.0 statistics and SPSS 18.0 modeler were used to conduct clustering analysis and association rule analysis on core drugs respectively.Result:1.General literature results:A total of 3083 articles were collected.After deleting the duplicates and screening them in detail,356 articles were finally included,including 151 clinical study literatures and 205 animal experimental study literatures.In 151 clinical study literatures,143 literatures include the complete composition and dosage of drug,and 45 literatures include classical formulas.In 205 animal experimental study literatures,125 literatures include the complete composition and dosage of drug,and 143 literatures include classical formulas.2.Results of clinical study literature for the treatment of AD:(1)A total of 170 formulas were collected,and 174 kinds of traditional Chinese medicine with the total frequency of 1842 were sorted out,including 34 core drugs with the cumulative frequency rate of 70.74%,including Acorus Gramineus,Rhizoma Chuanxiong,Radix Rehmanniae Preparata,Radix Polygalae,Poria,Radix Glycyrrhizae,Radix Astragali,Radix Angelicae Sinensis,Salvia Miltiorrhiza,Fructus Corni,Radix Polygoni Multifloris,Fructus Lycii,Herba Epimedii,Radix Codonopsis,Fructus Alpiniae Oxyphyllae,Rhizoma Atractylodis Macrocephalae,Radix ginseng,Semen Persicae,Rhizoma Dioscoreae,Radix Paeoniae Rubra,Herba Cistanches,Pericarpium Citri Reticulatae,Flos Carthami,Radix Curcumae,Hirudo,Fructus Schisandrae,Pheretima,Radix Morindae Officinalis,Rhizoma Polygonati,Arisaema Cum Bile,Radix Paeoniae Alba,Semen Zizyphi Spinosae,Rhizoma Pinelliae,Semen Cuscutae.Acorus Gramineus was used for 94 times.(2)The core drugs were consisting of 10 formulations,including tonic-deficiency,blood-activating and stasis-resolving,tranquilizing,cough-suppressing and panting-calming,astringent,orifice-opening,dampness-draining diuretic,heat-clearing,qi-relating,liver-pacifying and wind-extingushing drug.Among them,Tonic-deficiency drug(17 drugs),blood-activating and stasis-resolving drug(6 drugs),account for 67.65%of the 34 core drugs.(3)Four clustering formulas and 63 association rules were obtained by clustering analysis and association rule analysis of core drugs.(4)36 classical formulas with the total frequency of 73 were sorted out and 16 high-frequency classical formulas with a cumulative frequency rate of 72.60%,including Tongqiaohuoxue Decoction,Qifu Beverage,Dihuangyinzi,Huanglianjiedu Decoction,Xixin Decoction,Jinkuishenqi Pill,Didang Decoction,Buyanghuanwu Decoction,Yigan Powder,Yiqicongming Decoction,Ditan Decoction,Huanshaodan,Zhibai Dihuang Decoction,Taohong Siwu Decoction,Zuogui Pill,Danggui Shaoyao Powder.3.Results of animal experiment literature for the treatment of AD:(1)A total of 130 prescriptions were collected,and 122 kinds of traditional Chinese medicine with the total frequency of 1076 were sorted out,including 33 core drugs with the cumulative frequency rate of 70.54%,including Acorus Gramineus,Poria,Radix Polygalae,Rhizoma Chuanxiong,Radix Rehmanniae Preparata,Radix Glycyrrhizae,Radix Astragali,Radix Ginseng,Radix Angelicae Sinensis,Radix Ophiopogonis,Fructus Corni,Radix Paeoniae Alba,Rhizoma Zingiberis Recens,Radix Polygoni Multiflori,Rhizoma Atractylodis Macrocephalae,Herba Dendrobii,Herba Cistanches,Radix Aconiti Lateralis Preparata,Fructus Schizandrae,Herba Menthae,Radix Morindae Officinalis,Fructus Jujubae,Radix Salviae Miltiorrhizae,Cortec Moutan,Cortex Cinnamomi,Herba Epimedii,Radix Rehmanniae,Ramulus Cinnamomi,Rhizoma Pinelliae,Semen Persicae,Radix Codonopsis,Radix Paeoniae Rubra,Fructus Ligustri Lucidi.Acorus gramineus was used for 73 times.(2)The core drugs were consisting of 10 formulations,including the drug for tonic deficiency,blood-activating and stasis-resolving,exterior-releasing,heat-clearing,astringent,interior-warming,orifice-opening,dampness-draining diuretic,tranquilizing,cough-suppressing and panting-calming.Among them,Tonic deficiency drug(16 drugs),blood-activating and stasis-resolving drug(3 drugs),exterior-releasing drug(3 drugs)and heat-clearing drug(3 drugs),account for 75.76%of the 33 core drugs.(3)Four clustering formulas and 141 association rules were obtained by clustering analysis and association rule analysis of core drug.(4)43 classical formulas with the total frequency of 152 were sorted out and 14 high-frequency classical formulas with a cumulative frequency rate of 70.39%,including Dihuangyinzi,Xiaoyao powder,Huanglianjiedu decoction,Buyanghuanwu decoction,Kaixin powder,Heixiaoyao powder,Yangshou pellet,Dangguishaoyao Powder,Liuweidihuang pill,Xixin decoction,Danzhi Xiaoyao Powder,Ditan Decoction,Erzhi Pill,Qifu beverage.Conclusion:1.In clinical study or animal experimental study for the treatment of traditional Chinese medicine in AD,Acorus Gramineus is the most frequently used single drug.2.In clinical study for the treatment of AD,34 traditional Chinese medicines such as Acorus gramineus,Rhizoma Chuanxiong,Radix Rehmanniae Preparata,Radix Polygalae,Poria,Radix Glycyrrhizae,Radix Astragali,Radix Angelicae Sinensis,Salvia Miltiorrhiza,Fructus Corni and so on are the core drugs.In clinical study for the treatment of AD,16 classical formulas such as Tongqiaohuoxue Decoction,Qifu Beverage,Dihuangyinzi,Huanglianjiedu Decoction,Xixin Decoction,Jinkuishenqi Pill,Didang Decoction,Buyanghuanwu Decoction,Yigan Powder,Yiqicongming Decoction and so on are the core classical formulas.In animal experimental study for the treatment of AD,33 traditional Chinese medicines such as Acorus Gramineus,Poria,Radix Polygalae,Rhizoma Chuanxiong,Radix Rehmanniae Preparata,Radix Glycyrrhizae,Radix Astragali,Radix Ginseng,Radix Angelicae Sinensis,Radix Ophiopogonis and so on are the core drugs.In animal experimental study for the treatment of AD,14 classical formulas such as Dihuangyinzi,Xiaoyao Powder,Huanglianjiedu Decoction,Buyanghuanwu Decoction,Kaixin Powder,Heixiaoyao Powder,Yangshou Pellet,Dangguishaoyao Powder,Liuweidihuang Pill,Xixin Decoction and so on are the core classical formulas.3.In clinical study for the treatment of traditional Chinese medicine in AD,the core drugs mainly include the drugs for tonic deficiency,blood-activating and stasis-resolving.In animal experiment study for the treatment of traditional Chinese medicine in AD,the core drugs mainly include the drugs for tonic deficiency,blood-activating and stasis-resolving,exterior-releasing,heat-clearing.Tonifying the kidney and fortifying the spleen,tonifying qi and replenishing blood,activating blood and resolving stasis are the major direction in these two studies.
