Objective:To study the dynamic process of red blood cells carrying/releasing oxygen,and analyze the oxygen-carrying ability and membrane protein expression changes of red blood cells in different storage times.To provide scientific basis and guidance for improving the quality of blood preservation and the efficacy of clinical transfusion.Methods:To selective 10 case of elective orthopedic surgery,ASA?40???,aged 25?50years old,were subjected to normovolemic hemodilution before anesthesia,50 ml of whole blood was collected,and plasma was separated under closed conditions to prepare suspended red blood cells.The effective oxygen carrying capacity?Q?,P50,2,3-DPG,and Na+-K+-ATPase of red blood cells were measured at 1th,7th,14th,21th,and 28th days of storage,respectively,and erythrocyte morphology was observed by electron microscopy.EPB41,S1P protein expression levels were detected by Western Blot on 1th,7th,14th,21th and 28th days of storage.Results:Q value,P50,2,3-DPG and Na+-K+-ATP all decreased with the increase of storage time.Compared with the 1th day of storage,the Q value was significantly reduced on the 7th,14th,21th and 28th days of storage time?P<0.05?;the P50 was significantly reduced on the 14th,21th and 28th days of the storage time?P<0.05?;2,3-DPG concentration was significantly reduced on the 7th,14th,21th and 28th days of storage time?P<0.05?;Na+-K+-ATP concentration was significantly reduced on the 14th,21th,and 28th days of the storage time?P<0.05?.Compared with storage for 1th day,by the end of storage,the red blood cell Q value,P50,2,3-DPG,and Na+-K+-ATP decreased by36.36%,13.50%,94.09%,and 62.96%,respectively.Electron micrographs of erythrocytes showed that red blood cells were normal on the 1st days of storage time,and the red blood cells appeared as spinous processes on the 7th day of storage.By the end of storage,a large number of red blood cells lost their normal morphology.Protein EPB41,S1P all decreased with the increase of erythrocyte storage time.There was no significant difference in the first 14 days?P>0.05?,while the results of protein expression existed significant difference on 28th day and 21th day comparing with the 1th day?P<0.05?.Conclusion:Oxygen-carrying capacity and membrane protein expression of red blood cells both decrease during the preservation process.The decrease of oxygen-carrying capacity may be related to the inhibition of membrane protein expression.
