Objective Under the different concentration and adding time of L-arginine on suspended red blood cells of different inventory,to explore the influence of the preservation of red blood cells oxygen-carrying capacity,and to increase the ability of the preservation of red blood cells oxygen-carrying capacity.To explore for freezing red blood cells damage mechanism and protection strategy to provide important basis,dose,at the same time to fturiher clear to the erythrocyte function to realize quantitative infusion,improve the effect of clinical infusion of red blood cells to provide more theoretical basis,to explore a new type of erythrocyte preserve fluid add L-arginine formula,improve the quality of blood preservation,provide the experimental data.Methods1.Different concentrations of L-arginine effect on the ability of red blood cells oxygen-carrying capacity,preserved in suspended red blood cells of CPDA preservation solution respectively add L-arginine solution,the final concentration of 5mmol/L,10 mmol/L,50 mmol/L,in inventory period of 0 d,7 d,14 d,21 d,28 d,35 d detecting the change of the red blood cell oxygen affinity P50,2,3-DPG and the change of ATP and its influence to the red blood cell membrane.2.The effects of different L-arginine,adding time of red blood cells oxygen-carrying capacity,respectively before the frozen red blood cells on ice,frozen thawed MAP erythrocyte preserve preservation solution with L-arginine solution,the final concentration of 5 mmol/L,10 mmol/L,50 mmol/L,respectively in inventory period 0 d,7 d,14 d,21 d,28 d,35d,detected the frozen red blood cell oxygen affinity P50,frozen red blood cells,2,3-DPG and the change of ATP and its influence to the frozen red cell membrane.3.The methods of detection of the erythrocyte oxygen-carrying capacity:the hemoglobin oxygen-carrying/oxygen-releasing analyzer independently developed by the academy of military medical sciences was used to determine the oxygen dissociation curve and oxygen affinity P50 values of blood sanples at 0d,7d,14d,21d,28d,35d during different storage periods.The absorbance value was read by the enzyme label instrument,and the red blood cell oxygen carrying capacity was evaluated by measuring the concentration of 2,3-DPG and ATP in red blood cells by spectrophotometry.The effects of L-arginine on erythrocyte membranes were evaluated by determining the concentration of pH and free hemoglobin.Results1.With the prolongation of the inventory period,the oxygen affinity P50 of suspended red blood cells showed an overall decrease trend,especially in the beginning of the inventory period within 2 weeks,decreased by 15.6%in the inventory period within 14 days,then decreased slowly,and decreased by 25.4%on the 35d.At the end of the inventory period,its value was only 74.6%on the day of blood collection.The regression analysis of the experimental data showed that there was a linear correlation between P50 and inventory time.2.By the extension of the inventory period,frozen red blood cells are in the overall downward trend,the oxygen affinity of P50 and compared with suspended red blood cells,red blood cells in the inventory period after frozen at the same time points are in reducing the oxygen affinity of P50 change,carries on the regression analysis of experimental data,found that after frozen red blood cells of P50 and inventory time both are linearly related.3.The L-arginine group with a final concentration of 10mmol/L had the most significant effect on the improvement of oxygen affinity related index P50 of suspended red blood cells,and the difference between the two groups was statistically significant(P<0.05).P50 value of oxygen affinity related index P50 at the end of storage was still 82.5%on the day of blood collection.The L-arginine group with a final concentration of 5mmol/L had a certain increase in the oxygen affinity of suspended red blood cells,but the difference between the two groups was not statistically significant(P>0.05).However,the effect of L-arginine group with a final concentration of 50mmol/L on theoxygen affinity related index P50 of suspended red blood cells compared with the control group was not improved,but the oxygen affinity related index P50 was significantly decreased dxuring the inventory period.4.L-arginine increased the oxygen affinity P50 of red blood cells in frozen storage more than that of suspended red blood cells,and 1-arginine was added to the frozen thawed complex solution after the preservation and thawing of frozen red blood cells.Similarly,1-arginine with a final concentration of 10 mmol/L had the most significant effect on the improvement of oxygen affinity P50 in frozen red blood cells.5.As the storage period was prolonged,the concentrations of 2,3-DPG and ATP in the suspended red blood cells and the frozen preserved red blood cells were gradually consumed,while L-arginine had no significant effect on the increase of the concentrations of 2,3-DPG and ATP in the suspended red blood cells and the frozen preserved red blood cells,and there was no statistically significant difference from the control group without L-arginine.6.With the extension of the storage period,the free hemoglobin concentration of the suspended red blood cells and the frozen preserved red blood cells presented a gradually increasing trend,especially after 21 days of storage period,and the free hemoglobin concentration of the frozen preserved red blood cells was higher than that of the suspended red blood cells in the same storage period.L-arginine had a greater effect on the reduction of free hemoglobin in frozen erythrocytes than that in suspended erythrocytes,and L-arginine was added to the frozen thawed erythrocytes to have the most significant effect.L-arginine with a final concentration of 10mmol/L had the most obvious effect on the erythrocyte membrane.Conclusion1.With the prolongation of the storage period,the oxygen affinity P50 of the suspended red blood cells and the frozen red blood cells showed an overall decrease trend,and the oxygen carrying capacity of the red blood cells decreased continuously.The oxygen affinity P50 of the red blood cells showed a linear correlation with the storage time.2.With the prolongation of the storage period,the concentration of free hemoglobin in suspended red blood cells and frozen red blood cells presented a gradually increasing trend,the number of red blood cell destruction gradually increased,and the stability of the red blood cell membrane continuously decreased.3.The addition of L-arginine at an appropriate concentration during the storage period improved the oxygen affinity Pso of the suspended red blood cells and the stability of the erythrocyte membrane after frozen preservation and thawing,and the addition of L-arginine to the complex solution after frozen thawing was the most significant.
