Objective:By observing the efficacy of promoting blood circulation and removing blood stasis medicine,wind medicine,promoting blood circulation and removing blood stasis medicine with wind medicine on rats with ischemic stroke.To explore the therapeutic effect of promoting blood circulation and removing blood stasis medicine and wind medicine on ischemic stroke,and the synergistic effect and mechanism of promoting blood circulation and removing blood stasis medicine and wind medicine on the treatment of ischemic stroke,so as to provide new ideas for the clinical treatment of ischemic strokeMethods:1.Modeling and grouping:150 healthy adult male SD rats were randomly divided into sham-operation group,model group,promoting blood group,wind group and durg combination group,there were 30 rats in each group.Basis on Suture-occluded method building middle cerebral artery occlusion(MCAO)model in rats.After 24 hours of successful modeling,gastric gavage was started for 7 d.2.Texting indexs:The score of neural function was measured by the five-point method and the volume of cerebral infarction was measured by TTC,dry-wet weighing method to measure brain water content,Evans blue method to assess BBB permeability,HE staining to observe the pathological changes of brain tissue in rats,detecting the apoptosis of nerve cells in the hippocampus of brain tissue by TUNEL,westeron blot measure its expression levels of PI3K,Akt and CREB in brain tissuesResults:1.NSS:NSS in model group decreased significantly compared with that in sham-operation group(P<0.01);Compared with model group,NSS in each treatment group was lower than that in model group(P<0.05),and the NSS in durg combination group was the lowest(P<0.01).2.TTC staining was used to calculate the volume of cerebral infarction:brain tissue TTC staining showed that sham-operation group was uniformly red with no infarction,while model group showed obvious white infarction area(P<0.01).Compared with model group,the white cerebral infarction was significantly reduced in each treatment group(P<0.01),and the infarction volume was the smallest in durg combination group(P<0.01).3.Determination of brain water content by the dry-wet weighing method:the brain water content of rats in model group was significantly higher than that in sham-operation group(P<0.01);Compared with model group,the brain water content of each treatment group decreased(P<0.01),and the most significant decrease was in durg combination group(P<0.01).4.Evans blue method to evaluate blood-brain barrier permeability:the content of Evans blue in the brain tissues of rats in model group was significantly higher than that in sham-operation group(P<0.01);Compared with model group,the content of Evans blue in the brain tissues of the rats in each treatment group decreased(P<0.01),and the content of Evans blue decreased most significantly with durg combination group(,P<0.01).5.Histomorphological changes of rats:there were no morphological changes in the brain tissue of sham-operation group,while the cells in the brain tissue of model group were disordered and morphological destruction.Compared with model group,the morphological structure of brain tissue in each treatment group was improved,among which durg combination group had the best effect.6.Expression of apoptotic nerve cells in rat brain tissue:no apoptotic cells were observed in sham-operation group(P<0.01);Compared with sham-operation group,the apoptotic cells in model group increased significantly(P<0.01).Compared with model group,the apoptotic cells in each treatment group were significantly reduced(P<0.01),and the apoptosis of nerve cells in durg combination group was the least(P<0.01).7.Expression of PI3K,Akt and CREB in brain tissues of rats:a large number of PI3K,Akt and CREB proteins were observed in sham-operation group,which were significantly inhibited after modeling(P<0.01).After drug treatment,the expression of PI3K,Akt and CREB proteins was significantly increased in each treatment group(P<0.05),with the most significant effect in durg combination group(P<0.01).Conclusion:1.Both promoting blood circulation and removing blood stasis medicine,wind medicine,promoting can improve the NSS of MCAO model rats,reduce the volume of cerebral infarction,permeability of blood-brain barrier and the apoptosis of nerve cells.In addition,wind medicine has synergistic effect on the treatment of promoting blood circulation and removing blood stasis in ischemic stroke.2.The synergistic effect of wind medicine on the treatment of promoting blood circulation and removing blood stasis in ischemic stroke may be related to the PI3K/Akt signaling pathway.
