Objective: To explore the clinical application value of automatic multiplexed magnetic bead immunoassay for the level of common autoantibodies in Serum of patients with systemic lupus erythematosus(SLE),and to provide reference and reference for early diagnosis and timely treatment of clinicians.Methods: A total of 139 sera from confirmed SLE patients admitted to the Department of Rheumatology and Immunology of the First Affiliated Hospital of Bengbu Medical College were selected from December 2017 to December 2019,and the levels of common autoantibodies in Serum of the above patients were qualitatively and quantitatively determined using automatic multi-bead immunoassay;According to the matching of admission time,gender,age,and course of disease,the serum of 30 patients was collected and the levels of anti-ds DNA,anti-Sm and other major autoantibodies were measured by enzyme-linked immunosorbent assay(ELISA);An automatic multiplexed magnetic bead immunoassay was used to qualitatively and quantitatively detect autoantibodies in Serum of 33 healthy control groups with gender and age matching.At the same time,the general data and laboratory indicators of SLE patients were collected and sorted,and the two variables that met the normal distribution were statistically tested using the t test.The two variables that did not meet the normal distribution were tested by the rank sum test.Chi-square test was used for comparative analysis.The correlation between the two variables was analyzed by Spearman correlation,and the consistency of the two methods was compared by Kappa consistency analysis.Results: Automatic multiplexed magnetic bead immunoassay for 15 antinuclear antibodies in 139 patients with SLE,and 134 patients had positive results.The total positive rate was 96.4%,and the specificity was 97.0% compared with the healthy control group.The positive rates of anti-ds DNA,CHrom,Ribo-p,Sm,Sm RNP,RNP,RNP68,SSA,RNP-A,Ro60,Ro52,and SSB were 58.3% and 70.5% in SLE patients.,32.4%,41.0%,57.6%,55.4%,36.0%,49.6%,51.1%,56.1%,36.7%,19.4%,compared with the healthy group(P <0.05),Compared with healthy group,there was significant difference in Scl-70,Cen B,Jo-1 between healthy group(P > 0.05).There were no statistically significant differences in the positive rates of anti-ds DNA,Ro60,Ro52,SSB,and Ribo-p detected by the two methods of automated multiplexed magnetic bead immunoassay and ELISA(P> 0.05),while the positive rates of anti-Sm had statistical difference(P <0.05).The two methods have good consistency in detecting ds DNA,Ro60,SSB,and Ribo-p,and their Kappa values are 0.79,0.87,0.76,and 0.80,;the two methods have average consistency in detecting Sm and Ro52,and their Kappa values are 0.61,0.50.ESR and CRP in the chrom opositive group increased in the SLE patient group;WBC decreased;anti-ds DNA and Sm levels increased significantly(all P <0.05);Spearman correlation analysis showed that CHrom levels and ESR(r = 0.317,p = 0.000),CRP(r = 0.172,p <0.05),but no correlation with WBC(r =-0.156,p = 0.066),CHrom in serum anti-ds DNA negative group and anti-Sm negative group The positive rates were 53.4% and 54.9%.Automatic multiplexed bead Immunoassay full automation,detection of 1,300 indicators / hour,simultaneous detection of multiple indicators,continuous sample loading,the first report time of 45 minutes,supporting emergency department Detection.ELISA manual / semi-automatic,1 plate(96 samples):> 2 hours,single indicator detection,batch sample loading,the first report time is greater than 2 Hours,emergency testing is not supported.Conclusion: The sensitivity and specificity of the automatic multiplexed bead Immunoassay for Serum in patients with SLE are similar to the results of ELISA commonly used in clinical practice.The sensitivity of the serum anti-Sm antibody is higher than that of ELISA;the serum anti-ds DNA negative and anti-Sm-negative SLE patients have a certain reference value for their serum anti-CHrom antibodies;because automatic multiplexed bead Immunoassay can automatically,quickly and simultaneously quantify,and multi-flux detection of multiple antibodies,Worth promotion and application.
