Objective:To explore the effect of aerobic exercise on the levels of a utop ha gy-related markers and autophagy activation in mouse aorta,we let the mice undergo aerobic exercise and then detect the expression of mRNA and protein of autophagy-related factors in mice aorta.We hope to provide a theoretical basis for aerobic exercise improving atherosclerosis.Methods:Forty C57BL/6 male mice aged 8 weeks were randomly divided into quiet control group(n=20)and aerobic exercise group(n=20 10 for Real-Time PCR and 10 for Western-blot).The aerobic exercise group were subjected to swimming.On the first day,the mice swim for 10 minutes,then 10 minutes longer per day,until 60 minutes.After that the mice maintain this 60-minute exercise 5 days a week for 6 weeks.The quiet control group was free to move.The Thoracic and abdominal aorta of mice were isolated and detected after 6 weeks.Real-Time PCR was adopted to measure the mRNA levels of autophagy-related markers,including ULK1,Beclinl and LC3B.Western-blot was applied to measure the expression level of autophagy-related protein including the ratio of LC3B-II/LC3B-I and p62.The data were analyzed by independent sample t test.Results:Compared with the quiet control group,in the aerobic exercise group,the mRNA levels of ULK1,Beclinl and LC3B and the ratio of LC3B-II/LC3B-I were significantly increased(P<0.05),the protein degradation of p62 was significantly increased(P<0.05).Conclusion:Aerobic exercise can increase the expression of ULKI,Beclin1 and LC3B in the aorta,and then enhance the aortic autophagy activity.
