Lonicera Japonica Flos is a dry flower bud of Lonicera japonica Thunb.It is one of the most commonly used in Traditional Chinese Medicine(TCM),whose functions include heat detoxification and evacuation of wind heat.During the fight against COVID-19,Lonicera Japonica Flos played a crucial role,with prices reaching 280 yuan/kg.There are 98 species of Lonicera Linn in China,among which Lonicera maackii(Rupr.)Maxim is very similar to Lonicera Japonica Flos,Lonicera maackii is used instead of Lonicera Japonica Flos in a few areas.However,the efficacy of Lonicera maackii is not clear.If used instead of Lonicera Japonica Flos can't guarantee the efficacy,especially during the epidemic,it will be latent huge hidden trouble.At present,there are few studies on the identification of Lonicera Japonica Flos and Lonicera maackii.In this paper,the identification of the two is carried out by combining traditional and modern technology,the results are as follows:1.Through the observation of the characters and powder characteristics of L.Japonicae and L.maackii,it was found that there were a difference in the length of L.Japonicae and L.maackii in appearance,in which L.Japonicae was 2?3cm,and L.maackii was 1?2cm.There are some differences in powder characteristics such as glandular hairs,pollen grain surface glyph,thick-walled nonglandular hairs and thin-walled nonglandular hairs.Among them,the glandular hairs of L.maackii is very short,which is composed of 1?2 cells,the glandular hairs of L.Japonicae is longer,which is composed of 2?4cells;The length of thin-walled nonglandular hairs of L.Japonicae is much longer than that of L.maackii.2.Sixteen different batches of L.Japonicae and L.maackii were determined by HPLC,and the similarity was evaluated by traditional Chinese medicine chromatographic fingerprint similarity evaluation system(2004A version),and identified by cluster analysis.Results the HPLC fingerprints of L.Japonicae and L.maackii were established,the similarity between them was 0.972-0.994,and they could be completely distinguished by cluster analysis,indicating that the chemical composition of them was simi-lar,but the contents of effective components were different.HPLC fingerprint has high accuracy and specificity,which can be used to identify the authenticity of L.japonicae.3.The psbA-trn H gene sequences of L.Japonicae and L.maackii were determined by direct sequencing of PCR products,and download 7 sequences of psbA-trn H genes from GENBANK.All the sequences were compared by MEGA7.0,calculate the genetic distance between intraspecific and interspecific and the cluster tree of ML system was constructed.The results showed that the maximum intraspecific genetic distance(0.003)of L.Japonicae and L.maackii was much smaller than the interspecific minimum genetic distance(0.105),and there was obvious bar code distance between them.At the same time,the results of ML tree could clearly distinguish them.So the psbA-trn H sequence can be used to identify L.Japonicae and L.maackii.4.Several batches of L.Japonicae and L.maackii from different years and different producing areas were collected,and the best measurement conditions were determined by orthogonal test under SuperNose-14 electronic nose system,then the sample odor information was measured.Principal component analysis(PCA)and linear discriminant analysis(LDA)were used to analyze the sample odor information,and the identification models of authenticity,origin and storage year were established respectively.The results show that LDA recognition model can clearly distinguish between L.Japonicae and L.maackii,different producing areas and different storage years.Making use of the special smell of L.Japonicae,the electronic nose was used to distinguish L.Japonicae in detail,and the distinguishing effect was remarkable,which provided a new idea for the identification of L.Japonicae in the future.
