Inonotus obliquus(Fr.)Pilat is a precious and rare medicinal fungus,known to be effective for the prevention and treatment of such incurable diseases as cancer and diabetes.In addition,Drosophila melanogaster s an efficient model with such advantages as short life cycle and low cost for the bioactivity evaluation of traditional Chinese medicine.In this study,the effects of I.obliquus polysaccharide on gut immunity and insulin metabolism of D.melanogaster were investigated through toxic reagent-induced gut damage and intestinal stem cell(ISC)tumor models and high sucrose diet(HSD)-induced and insulin signaling pathway-mutated diabetes models by analyzing survival rate,number of dead cells,reactive oxygen species(ROS),Diptericin expression,ISC proliferation and differentiation,percentage of flies with tumor and growth indices such as pupa size and adult body weight.1.The polysaccharide ameliorated toxic reagent-induced Drosophila survival rate decrease and excessive gut epithelial cell death and alleviate not only the enhanced ROS and Diptericin levels and the excessive ISC proliferation and differentiation due to the gut damage but also the ISC tumorigenesis induced by the combination of tissue stress and ISC differentiation loss.2.The polysaccharide was proved to ameliorate the reductions at growth indices in HSD-fed Drosophila and chico mutant but not in S6k mutant.This study provides a theoretical basis for the further research on the detailed mechanism of I.obliquus polysaccharide.
