Study On Extraction Of Inonotus Obliquus Polysaccharide And The Activity

Inonotus obliquus has become a kind of new edible and medicinal fungi since the 1990's.It contains rich biological activity components. Up to date, not much research was focused on its immunological activity abroad. We have not found reports of such aspects in China yet. In this article Inonotus obliquus were served as experiment materials , and coarse polysaccharides was extracted from them. The purpose was to study the effect of different factors on the yield and purity of Inonotus obliquus polysaccharide and find the best extracting condition by means of orthogonal.To study the non-specific immunological function of Inonotus obliquus on normal and immuno-suppression by using carbon particle clearance test. In order to investigate the cellular immune function of Inonotus obliquus on normal and immuno-suppression , by using the lymphocyte proliferation test. To study the specific humoraimmunological effect of Inonotus Obliquus on normal and immuno-suppression by using the hemolysin test.In this article moisture content, protein content, cinerationg content, fat content, trace element and heavy metal are tested. And these facts sustained the theory of research and development.The results of test were as follows:(1) This experiment used the hot water immersion formulation to extract the Inonotus obliquus polyszcchride. The single factor experiment was based on those 4 craft parameters and designed the orthogonal experiment in this foundation, .And the proposing extracting Inonotus obliquus polysaccharide best condition was the best soaks raises temperature was 100℃, that the best material hydraulic modulus was 1:25,and that the best soaks raises time was 4 hours. And in this condition the Inonotus obliquus polysaccharide yield was 1.841%.(2) Coarse polysaccharides of Inonotus obliquus can obviously increased spleen index and phagocytic rate of macrophage and strengthen non-specific immunological level of organism.(3) They can noticeably stimulate lymphocytes. Therefore, experiment indicated that coarse polysaccharide of Inonotus obliquus was a stimulating agent of lymphocytes which can promote immunological level of organism cells.(4) They can obviously increase the content of haemolysin which circulated antibody and could cause organism to obtain the ability to produce antibody which can finally increase the immunological level of body fluid.(5) When we treated coarse polysaccharides and Cyclophosphamide at the optimal dose, coarse polysaccharides of Inonotus obliquus can weaken and even completely get rid of low immunological function caused by tumor chemotherapy and finally modify the low organism immunity into the normal level.(6) There were beneficial trace element such as Mn and Se. And the heavy metal contents were far-off less than the trade standard.Therefore, coarse polysaccharide of Inonotus obliquus is a good immune-strenthening agent. This study provides theory basis for developing protective medi-cine.
