Objective:(1)To explore the expression of TGF?R2 protein and mismatch repair(MMR)protein in colorectal cancer(CRC)tissues;(2)Explore TGF?R2 in CRC and adjacent tissues and in microsatellite instability low-frequency(MSI-L)/microsatellite stability(MSS)group and microsatellite instability high-frequency,MSI-H)[6]Expression in the group;(3)To explore the relationship between the expression level of TGF?R2 in CRC and microsatellite instability(MSI)[6],and the correlation between the two and clinical pathological parameters.Method:(1)The multi-fluorescence PCR capillary electrophoresis and immunohistochemistry(IHC)En Vison two-step method was used to detect the MSI status of 184 cases of CRC tissue.(2)The real-time fluorescence quantitative PCR technique was used to detect the relative expression of TGF?R2 in 184 CRC tissues and adjacent tissues,as well as in MSI-L/MSS group and MSI-H group.(3)Statistical analysis:All data should be statistically analyzed using SPSS 26 statistical software.The Kappa test was used to test the consistency of the two test methods;the 2 test was used to test the correlation of the two classification data;the rank sum test was used to compare the means.P<0.05 has significant statistical difference.Result:(1)The expression level of TGFPR2 in CRC tissue is lower than that in adjacent tissues(P<0.05).(2)The relationship between the relative expression of TGF?R2 and the clinicopathological parameters of patients with CRC:the relative expression of TGF?R2 mRNA in colon cancer tissue is higher than that in rectal cancer;the relative expression of TGF?R2 mRNA in cancer tissue that has not metastasized to lymph nodes is Cancer tissue that is higher than lymph node metastasis;the relative expression of TGF?R2 mRNA in TNM stage I-II cancer tissue is higher than that in stage ?-? cancer tissue;TGFPR2 mRNA in cancer tissue that has not metastasized distantly The relative expression level was higher than that of cancer tissues with distant metastasis.The difference was statistically significant(P<0.05).It was not related to the patient's sex,age,tumor maximum diameter,degree of differentiation,histopathological type,tumor infiltration depth,and presence or absence of tumor thrombus in the vessel.The difference was not statistically significant(P>0.05).(3)The relationship between MSI and clinical pathological parameters of CRC patients:MSI-H tumors mostly occur in colon,non-lymph node metastasis,and stage?-? CRC patients.MSI-L/MSS tumors are mostly high-and medium-differentiated,with differences It is statistically significant(P<0.05).It was not related to the patient's gender,age,tumor maximum diameter,pathological type,tumor infiltration depth,presence or absence of tumor thrombus in the vessel,and whether the tumor had distant metastasis.(4)In CRC tissue,the relative expression of TGF?R2 mRNA in MSI-H group was significantly higher than that in MSI-L/MSS group(P<0.05),suggesting that the deletion of TGF?R2 gene was positively correlated with MSI-H.(5)The expressions of MSI and TGF?R2 are consistent and synergistic in the location of tumor occurrence,lymph node metastasis,and TNM staging.Conclusion:The expression of TGF?R2 in colorectal cancer is highly positively correlated with MSI.The two are synergistic in clinical pathological parameters such as tumor site,lymph node metastasis,and TNM staging.TGF?R2 is expected to become a potential colorectal cancer occurrence,development and treatment prognostic biomarkers for efficacy.
