| ObjectiveObservation of Dajianzhong Decoction and its decomposed formula on inflammatory cytokines IL-6,IL-8,TNF-α,serum gastrin(GAS)and gastric tissue vascular endothelial growth factor(VEGF)in rats with gastric ulcer The effects of related indexes,such as the protective effect of Dajianzhong Decoction and its decomposed prescriptions on gastriculcer and its related mechanisms were explored,and the compatibility of different drug groups in the treatment of gastric ulcer was explained.MethodsSixty SD rats,half male and half female,were randomly divided into blank group,model group,positive drug group,original prescription group,tonic group and scattered cold group,10 rats in each group.Except the blank group,the other five groups were established with rhubarb bitter cold and diarrhea method,hunger and satiety method combined with restraint water immersion stress method to establish spleen and stomach deficiency cold gastric ulcer model.The serum levels of IL-6,IL-8,TNF-α,GAS and gastric tissue VEGF were detected by ELISA method.The pathological changes of gastric tissue were observed by light microscope.SPSS22.0 statistical software was used to analyze and process the obtained data.Result1.In the process of spleen and stomach deficiency,the model rats gradually lose their energy,and even become sluggish,lazy,tired and tired,eat less,lose weight,lose weight,thin stools or mucus,and perianal fouling.The hair was dry and arrogant,and he could not insist on swimming and other performances.The macroscopic symptom scores of spleen and stomach deficiency were all above 5 points(P<0.01),suggesting that themodel of spleen and stomach deficiency due to rhubarb bitter cold and diarrhea was successfully established.2.Pathological morphological observation: Compared with the rats in theblank group,the gastric mucosa of the model group showed defects in the mucosal epithelium,edema in the submucosa,vascular congestion,bleeding tendency,and a large number of inflammatory cell infiltration on the mucosal surface;Compared with the model group,the pathological changes in each administration group were alleviated,and the original prescription group had the most obvious reduction.3.Ulcer index: Compared with the blank group,the ulcer index of the rats in the model group was significantly increased(P<0.01);compared with the model group,the ulcer index of the rats in each administration group was significantly reduced(P<0.01);Compared with the original prescription group,there was a significant difference between the tonic group and the scattered cold group(P<0.01),and the positive drug group had no statistical difference(P>0.05).4.Serum TNF-α,IL-6 and IL-8 levels: Compared with the blank group,the serum levels of TNF-α,IL-6 and IL-8 in the model group weresignificantly increased(P<0.01);Compared with the control group,the serum levels of TNF-α,IL-6,and IL-8 in each administration group were significantly reduced(P<0.01).Compared with the original prescription group,there was a significant difference between the positive drug group and the tonic group.(P<0.01),there was no difference in the scattered cold group(P>0.05).5.Serum GAS level: Compared with the blank group,the serum GAS of the model group was significantly increased,with significant differences(P<0.01);compared with the model group,the serum GAS of each administration group was significantly reduced,with differences.Significant difference(P<0.01);compared with the original prescription group,there were significant differences in the positive drug group,tonic group,and scattered cold group(P<0.01).6.VEGF level in gastric tissue: Compared with the blank group,the VEGF level in the gastric tissue of the model group rats was increased(P<0.05);compared with the model group,the VEGF level in the original prescription group and the tonic group was significantly increased,with a difference.Significant(P<0.01),the positive drug group increased(P<0.05),while the Sanhan group had no significant difference(P>0.05);compared with the original prescription group,there was no significant difference in the deficiency group(P>0.05),there was a difference in the positive drug group(P<0.05),and there was a significant difference in the scattered cold group(P<0.01).Conclusion:1.Dajianzhong Decoction and its disassembled prescriptions have different degrees of anti-gastric ulcer effect,but the overall control effectof the original prescription group is better than that of the Buxu groupand Sanhan group.2.In the decoction of Dajianzhong Decoction,the Sanhan group prefers to reduce the levels of inflammatory factors TNF-α,IL-6,and IL-8to suppress the inflammatory response to the gastric mucosa in order toexert its anti-ulcer effect;The deficiency group tends to increase the VEGF level,improve vascular permeability,promote the formation of granulation tissue and new blood vessels,and enhance the defense functionof the gastric mucosa.3.Dajianzhong Decoction has obvious prevention and cure effect onspleen and stomach deficiency cold gastric ulcer model rats,and its mechanism may be related to lowering serum inflammatory factors TNF-α,IL-6,IL-8 and reducing serum GAS levels.Associated with elevated gastric tissue VEGF levels. |
