Objective: By detecting the expression levels of S100A12 protein and PREX1 protein in synovial tissue of patients with rheumatoid arthritis(RA),osteoarthritis(OA)and Trauma,and the expression levels of S100A12 protein and PREX1 protein in serum of patients with RA,OA and healthy controls(HCS),and the expression level of S100A12 protein in the joint fluid of RA and OA patients,To investigate the diagnostic value of S100A12 and PREX1 proteins for RA and their correlation with clinical and laboratory indicators in RA.Comparison of the expression levels of S100A12 and PREX1 proteins in serum of patients with RA and rheumatoid arthritis-Interstitial lung disease(RA-ILD).Methods: The expression of S100A12 protein and PREX1 protein in synovial tissue of RA group(n=6),OA group(n=6)and NCS group(n=6)were determined by immunohistochemical(IHC).The protein content of S100A12 in the joint fluid of RA group(n=10)and OA group(n=10)and the expression levels of S100A12 protein and PREX1 protein in serum of RA group(n=56),RA-ILD group(n=28),OA group(n=45),and HCS group(n=40)were measured by Enzyme linked immunosorbent assay(ELISA).Determination of RA,RA-ILD the relevant laboratory indicators of the patients,and the collection of clinical data of the patients.the clinical value of S100A12 and PREX1 proteins in RA,RA-ILD patients was analyzed by statistical methods.Results: 1.The expression level of S100A12 protein in synovial tissue of RA patients was higher than that of OA group and NCS group.The expression level of S100A12 in the joint fluid of RA patients was significantly higher than that in OA group.2.Serological experiments showed that serum S100A12 protein and PREX1 protein levels of RA patients were significantly higher than those of OA group and HCS group,and the difference between OA group and HCS group was also statistically significant.The expression level of S100A12 protein in serum of RA-ILD patients was significantly higher than that of RA patients.Serum S100A12 protein level of RA patients was correlated with DAS28 score(r=0.497,p<0.01),28-TJC(r= 0.530,p<0.01),28-SJC(r=0.333,p<0.01)and RF-Ig M(r=0.423,p<0.05).The levels of PREX1 protein in serum with RA patients were not associated with relevant laboratory indicators.Multiple linear regression with DAS28 score as the dependent variable showed that the effect of S100A12 protein on DAS28 score was weaker than ESR,but better than CRP.According to ROC curve analysis,the sensitivity and specificity of S100A12 protein in RA diagnosis were 89.3% and 70.0% respectively.The positive rate of S100A12 was 89.2%(50/56)in RA group,53.3%(24/45)in OA group,and 30.0%(12/40)in HCS group.The sensitivity and specificity of PREX1 in RA diagnosis were 100% and 92.5% respectively.The positive rate of PREX1 protein in RA group was 100%(56/56),the positive rate of PREX1 protein in OA group was 22.2%(10/45),and the positive rate of PREX1 protein in HCS group was 7.5%(3/40).Conclusion: S100A12 and PREX1 proteins may be involved in the occurrence of RA,and may be a new biological marker for the diagnosis of RA,which has certain potential in the diagnosis of RA and the monitoring of disease activity.S100A12 protein may have a relationship with the ILD associated with RA.
