Objective: To investigate the association between telomerase gene(TERT)single nucleotide polymorphisms rs 10078761,rs 12696304,rs 2853669,rs 16847897,rs 2736100,rs 10069690 and benign prostatic hyperplasia(BPH).Methods: After obtaining informed consent,a total of 150 BPH patients and 150 normal elderly men who received treatment or health examination at the Second Hospital of Lanzhou University were included in the study.The remaining specimens of all subjects after normal clinical examination were collected and stored in a-80? refrigerator.The age,past medical history,height,weight,mean arterial pressure,International Prostate Symptom Score(IPSS),quality of life(QOL)score,overactive bladder symptom score(OABSS),peak urinary flow rate(Qmax),prostate volume(PV),postvoid residual urine volume(PVR),serum prostate specific antigen(PSA),pathological findings,and the presence or absence of recurrent hematuria,recurrent urinary tract infections,acute urinary retention,bladder stones and renal dysfunction were also recorded in the BPH patient group.For normal elderly male subjects,only age data were collected for statistical school positivity.DNA was extracted from the samples,and the polymorphic sites were detected by ligase detection reaction(LDR)+ multiplex PCR.Multiple linear regression and logistic regression were used to statistically analyze the detection results.Pearson correlation was used to analyze whether the target population was in Hardy-Weinberg equilibrium.Results: The population included in the experiment conformed to the Hardy-Weinberg equilibrium and was a stably inherited population.Rs 10078761,rs 12696304,rs 2853669,rs 16847897,rs 2736100,and rs 10069690 were not independently associated with the risk of clinical BPH by unit point analysis.However,a total of four haplotypes(TCTGGT,TCTGTC,TGCCTC,and TGTGTC)were found to be associated with the risk of clinical BPH by haplotype analysis.The independent population undergoing pathological examination conformed to Hardy-Weinberg equilibrium,and rs 2853669 was a factor associated with smooth muscle hyperplasia.In addition,rs 2736100,rs 10078761,and rs 10069690 are associated with clinical progression of BPH,and these three SNPs are in linkage disequilibrium.Conclusion: Telomerase is an important enzyme involved in cell proliferation,and the polymorphisms rs 2853669,rs 2736100,rs 10078761,rs 10069690 and some haplotypes of TERT gene in the population are associated with the clinical progression,pathological type and risk of clinical BPH.The C allele of rs 2853669 is an independent risk factor for smooth muscle-type hyperplasia.Alleles linked to Rs 2736100,rs 10078761,and rs 10069690 together increase the clinical progression of BPH.Four haplotypes,TCTGGT,TCTGTC,TGCCTC,TGTGTC(allelic order: rs 10069690,rs 2736100,rs 16847897,rs 2853669,rs 12696304,rs 10078761),were associated with clinical BPH risk.In summary,TERT gene polymorphism plays an important role in the occurrence and development of BPH...
