| ObjectiveTo establish a model of corticosteroid-induced high intraocular pressure in rats,and to observe the changes of intraocular pressure,trabecular meshwork and retina in the rats by intragastric administration of qidengmingmu capsule suspension,as well as to explore the mechanism of Qidengmingmu Capsule on reducing intraocular pressure and protecting optic nerve in rats with high intraocular pressure.MethodsThe rats were treated with 0.1% dexamethasone sodium phosphate eye drops three times a day for 36 days.55 male SD rats were randomly divided into 5 groups:low dose group,high dose group,acetazolamide group,high intraocular pressure model group,and a blank control group,11 rats each.The rats were administed with the suspension once a day for one month,and the general state and intraocular pressure of the right eye were observed.One month later,the rats were killed.Paraffin sections of the right eye were taken for histological measurement.TUNEL staining was used to detect the number of apoptotic cells.Flow cytometry,WST-1 method and TBA method were also used to measure the ROS level,SOD activity and MDA content in retina of the rats.Result1.During the preparation of corticosteroid-induced intraocular hypertension model,the right intraocular pressure of 44 rats in dexamethasone group reached more than 20 mmhg,and the success rate of the model was 91%.2.Compared with the high IOP model group,the right IOP in the low and high dose group of qidiangmingmu capsule decreased significantly;compared with the high IOP model group,the right IOP in the acetazolamide group decreased significantly;the right IOP in the low and high dose group of qidiangmingmu capsule was higher than that in the acetazolamide group and the difference was statisticallysignificant.3.The trabecular meshwork thickness of the right eye of dexamethasone-induced high intraocular pressure rats in each group were thinner than that of the blank control group.4.The number of trabecular meshwork cells in the right eye of rats with intraocular hypertension induced by dexamethasone was significantly reduced compared with the blank control group.There was no significant difference in the number of trabecular meshwork cells in the right eye between the Qi Deng Ming Mu capsule group and the high intraocular pressure model control group;the number of trabecular meshwork cells in the acetazolamide group was significantly higher than that in the high intraocular pressure model group.5.The thickness of retinal ganglion cells in the right eye of rats with intraocular hypertension induced by dexamethasone had a tendency of thinning in varying degrees,and there was no significant difference compared with the blank control group.6.Compared with the blank control group,the number of retinal ganglion cells in the right eye of rats with intraocular hypertension induced by dexamethasone decreased significantly.After the administration of gavage,the number of RGC in the low,high dose group and the acetazolamide group was significantly higher than that in the model group.7.The number of apoptotic cells in the right eyeball of rats with intraocular hypertension induced by dexamethasone was significantly increased compared with the blank control group.After intragastric administration,the number of apoptotic cells in the eyeball tissue of the low and high dose group was significantly less than that of the high IOP model group;there was no significant difference between the number of apoptotic cells in the eyeball tissue of the acetazolamide group and the high IOP model group.8.The activity of ROS in the right retina of rats with intraocular hypertension induced by dexamethasone was significantly higher than that of the control group.After gavage,the activity of ROS in the right retina of the high dose group andacetazolamide group was lower than that of high IOP model group.9.The activity of SOD in the right retina of rats with intraocular hypertension induced by dexamethasone was significantly lower than that of the control group.After gavage,SOD activity in the right retina of the high dose group was significantly higher than that of high IOP group.10.The content of MDA in the right retina of rats with intraocular hypertension induced by dexamethasone was significantly higher than that of the control group.After gavage,the MDA content in the right retina of the high dose group was significantly lower than that of the high IOP model group.Conclusion1.Using dexamethasone sodium phosphate eye drops to prepare rat model of high intraocular pressure is a successful non-invasive way,and the price is low.2.The number of apoptosis cells,the thickness of trabecular meshwork,the number of trabecular meshwork cells,the number of retinal ganglion cells,the activity of ROS,the activity of SOD and the content of MDA in the retina of the high intraocular pressure rats induced by steroid increased significantly.In addition,the thickness of the retinal ganglion cell layer also shows a trend of thinning.3.Qidengmingmu capsules can reduce intraocular pressure in steroid hormone-induced high intraocular pressure rats.4.Qi Deng Ming Mu capsule can resist the apoptosis of eyeball cells induced by corticosteroids in rats with high intraocular pressure.5.Qilianmingmu capsule can inhibit the activity of ROS,enhance the activity of SOD and reduce the content of MDA in the retina of;steroid-induced high intraocular pressure rats.It can reduce the apoptosis of RGC and protect the optic nerve by resisting the oxidative stress of retina. |
