Objective:This study examined the methylation status of FOXD3 gene in renal cancer tissues using related experimental methods.In addition,combined with clinical data,the possible mechanism of the role of FOXD3gene in the development of renal cell carcinoma(RCC)was explored.Methods:1.59 cases of RCC postoperative cancer tissues and corresponding normal tissues collected from the Fourth Hospital of Hebei Medical University were examined from January 2014 to December 2018.The methylation status of the FOXD3 promoter region in the tissues was detected by methylation-specific polymerase chain reaction.Results combined with clinical data was analyzed using statistical methods.2.The relative expression of FOXD3 m RNA in cancer tissues and normal kidney tissues of postoperative RCC from 59 patients was detected using Quantitative Real-time PCR(RT-q PCR)technology.The results were analyzed with relevant statistical data and clinical data.3.59 cases of RCC were divided into methylated RCC group and unmethylation RCC group according to whether methylation occurred.The differences in the relative m RNA expression levels between the two groups were compared to preliminary discuss whether the decrease in the relative m RNA expression level of FOXD3 gene in RCC tissues was related to the occurrence of methylation.4.SPSS 21.0 statistical software was used for the statistical analysis of the experiment data in this study.Results:1. HE staining resultsHE staining was used to examine if 59 tissue specimens were RCC or normal tissues.The results show there was no cancer cell infiltration found in normal tissues.2. MSP resultsThe incidence of methylation of FOXD3 gene in 59 normal kidney tissues and 59 RCC tissues was 32%(19/59)and 48%(28/59),respectively.The positive rate of methylation in RCC tissues was significantly higher than that in normal tissue specimens.(?~2=9.936842,P=0.001620)The analysis between FOXD3 methylation and the properties of patients.There were no significant differences between the methylation frequency of FOXD3and the properties of patients(such as age,gender,the size of tumors and cancer stage)between RCC tissues and normal kidney tissues(P>0.05).3. RT-q PCR resultsThe m RNA expression of FOXD3 in RCC tissues was significantly lower than normal kidney tissues(i.e.,0.583109(1.553664)and 1.00(0.01),respectively),showing there is a significant difference between RCC tissues and normal kidney tissues(P<0.05).Similar with the above MSP results,for both RCC tissues and normal kidney tissues,there is also no significant difference between the m RNA expression of FOXD3 and the characters of patients such as gender,age,the size of tumors and cancer stage)(P>0.05).Conclusions:1.The abnormal methylation of FOXD3 gene promoter region in RCC tissues may be related to the occurrence of RCC but not related with the characters of patients such as gender,age,the size of tumors and cancer stage.2.The decrease of the relative expression of FOXD3 gene in RCC tissues may be related to the occurrence of RCC but not related to the characters of patients such as gender,age,the size of tumors and cancer stage.3. The reason for the decrease of FOXD3 m RNA expression in RCC tissues may be due to the abnormal methylation of its promoter region.
