Objective Western blot analysis was performed on P65 of synovial fibroblast nf-kappab signaling pathway.In addition,the intervention factors HMGB1,IL-1 beta and HMGB1-IL-1complex were used to stimulate the 4 groups of experimental groups.To observe the amount of P65 production and explain the intrinsic relationship between HMGB1-IL-1 complex and NF-?B signaling pathway.Methods(1)Rheumatoid arthritis synovial fibroblast(RASFs)isolation,culture and identification: non-selective selection of 20 RA patients,digestive enzyme culture method and tissue block culture method to obtain synovial tissue.The culture samples were identified by optical microscope and Vimentin immunocytochemical staining,and whether they were RASFs was observed and the culture mode of RASFs was discussed.(2)the effect of HMGB1-IL-1 complex on kappa B signaling pathway(P65)of RASFs: Western blot analysis was conducted to detect the changes in the content of P65 in the nucleus of RASFs,and it was found that whether HMGB1-IL-1 beta could promote or inhibit kappa B signaling pathway.(3)statistical methods: all data were processed by SPSS 23.0 statistical software package for statistical processing.The measurement data of random,independent,normal(tested for normality)and homogeneity of variance(tested for homogeneity of variance)are expressed as mean ± standard deviation(X ± s).One-way analysis of variance was used to compare the mean of multiple groups.The snk-q test was used to compare the mean values of multiple samples in pairs.P < 0.05 means the difference is statistically significant.Results 1.Primary cells were cultured by digestive enzyme method and tissue block culture method.There was no significant difference between the two methods.2.High mobility group protein 1,interleukin 1 beta,and HMGB1-IL-1 complex intervention RASFs after 48 h,Western blot expression showed high mobility group protein 1-interleukin 1 beta complex intervention after 48 h of rheumatoid arthritis synovial fibroblasts p65 most abundant,there was no significantly difference.Conclusion Under experimental conditions in this study,the HMGB1-IL-1 complex can lead to pathological changes in RASFs and play an important role in the course of RA.TheHMGB1-IL-1 complex had a positive effect on the NF-kappa B signaling pathway of synovial fibroblast in ra,and was stronger than that in the independent group and the high migration rate group.
