BackgroundRheumatoid arthritis is a mutilation autoimmune disease. Cytokines play important effects on enlarging immunologic derangement and joints destroy. IL-1 and TNF-αare representative. In recent years, it s found that the high mobility group box protein 1 (HMGB1) involved in the pathogensis.ObjectiveIn the study, the experimental objects were CIA rats made by collagen-induced. To explore the role of HMGB 1 in arthritis and the antagonism of ethyl pyruvate to it by observing the amelioration of the arthritis and the HMGB1 expression of synovial tissue in CIA rat with ethyl pyruvate intervention.Methods1. Wistar rats were raised, received two times mixed liquor of cattleⅡcollagen,Freund's complete adjuvant (CFA) and Bacillus Calmette-Guerin (BCG) vaccine by endermic injection. CIA rats were made.2.24 CIA rats were divided to 2 groups randomly, and received ethyl pyruvate treatment or other kinds control drugs respectively. There are 12 CIA rats in normal control group. Since the third week, CIA rats have been induced by intraperitoneal injection of 40mg/kg ethyl pyruvate or physiological saline every other day. Then 6 rats in every group were killed after the 3th and 6th week.3. Observe the change of arthritis index, arthritis swelling degree, and appearance of imageology and pathology. Their synovium and joints were collected for observing the expression of HMGB 1 by immunohistochemistry and for analyzing the intensity of expression by IPP.Results1. Arthritis index, arthritis swelling degree, and appearance of imageology and pathology are labels,75 rats who had received mixed liquor of cattleⅡcollagen, CFA and BCG happened symmetry polyarthritis, achievement ratio of model is 77.14%;2. The evidences include the data of arthritis index and arthritis swelling degree by analysis of variance and the appearance of imageology and pathology. These confirmed that comparing with the control groups the joints were damaged obviously worse in experimental groups, while the arthritis of groups with intervention were inhibited in some extent.3. And at those two points, the HMGB1 expressions of HMGB1 antagon group are significantly higher than that of normal control (p<0.05). Additionally, there is statistical significance (p<0.05) about considerable augmentation of HMGB1 expression at placebo group. Furthermore, comparing the degressions of HMGB1 expression among three groups, the HMGB1 antagon group gets obvious descent and has statistical significance (p<0.05).Conclusions1. Ethyl pyruvate can improve the arthritis symptom, lower synovial inflammation and destruction of bone of collagen-induced arthritis rats.2. The results authenticated that HMGB1 induced inflammation in the synovial tissue of CIA rats, and provided the rationale that HMBG1 could be the target of curing RA.3. The presumption was that Ethyl pyruvate had the antagonism to HMGB1, which might provide a new therapeutic tool for patients with RA.
