Objective: To detect the gene expression of various subtypes of melanoma-associated antigen A(MAGE-A)in oral squamous cell carcinoma(OSCC)and precancerous lesions,to screen the main pathogenic factors that promote their carcinogenesis and metastasis,and to explore them as molecular markers for carcinogenesis and prognosis of oral squamous cell carcinoma(OSCC)and precancerous lesions.To study the role of MAGE-A in the carcinogenesis of oral precancerous lesions,lymph node metastasis and tumor invasion of OSCC,and to reveal its biological mechanism.The purpose of this study is to provide a theoretical basis for the clinical detection of the malignant potential of oral precancerous lesions and the early diagnosis,early intervention and prognosis of OSCC.Methods: The expression of MAGE-A1,A2,A3,A4,A5,A6,A8,A9,A10,A11,A12 in OSCC and precancerous lesions was detected by real-time fluorescence quantitative PCR(q PCR).The expression of MAGE-A subtypes in oral precancerous lesions with different pathological types of dysplasia and the relationship between MAGE-A subtypes and clinicopathological parameters were compared.The interaction among subtypes of MAGE-A was also discussed.Results: 1.MAGE-A m RNA was expressed in 18 cases of OSCC and 34 cases of precancerous lesions.The expression levels of MAGE-A1,A5,A6,A8,A9,A10,A11,A12 in 18 cases of OSCC were significantly higher than those in corresponding precancerous lesions(P<0.05).2.In 34 cases of oral precancerous lesions with different pathological types of dysplasia,the expression of MAGE-A m RNA in oral precancerous lesions of MAGE-A1,A2,A3,A4,A5,A6,A8,A9,A10,A11 was higher than that in mild epithelial dysplasia(P<0.05).The expression of MAGE-A1,A2,A3,A4,A5 in severe epithelial dysplasia was higher than that in moderate epithelial dysplasia(P<0.05),and the expression of MAGE-A6,A9,A12 in moderate epithelial dysplasia was higher than that in mild epithelial dysplasia(P<0.05).3.MAGE-A m RNA level was not related to sex,age,tumor size and tumor location in 18 patients with OSCC(P>0.05),m RNA levels of MAGE-A1,A5,A6,A8,A9,A10,A11 subtypes were related to TNM stage(P<0.05),and m RNA levels of MAGE-A1,A5,A6,A8,A9,A10,A11,A12 subtypes were significantly related to lymph node metastasis(P<0.05).4.There was a significant positive correlation among multiple MAGE-A subtypes in OSCC tissues(P<0.05): MAGE-A1 with MAGE-A2,A5,A6,A8,A9,A10,A11,A12;MAGE-A2 with MAGE-A1,A3,A4,A5;MAGE-A3 with MAGE-A2,A4,A5,A11;MAGE-A4 with MAGE-A2,A3,A5,A11;MAGE-A5 with MAGE-A1,A2,A3,A4,A6,A8,A9,A10,A11;MAGE-A6 with MAGE-A1,A5,A8,A9,A10,A11,A12;MAGE-A8 with MAGE-A1,A5,A6,A9,A10,A11,A12;MAGE-A9 with MAGE-A1,A5,A6,A8,A10,A11,A12;MAGE-A10 with MAGE-A1,A5,A6,A8,A9,A11,A12;MAGE-A11 with MAGE-A1,A3,A4,A5,A6,A8,A9,A10;MAGE-A12 with MAGE-A6,A8,A9,A10.Conclusion: 1.MAGE-A gene was expressed in OSCC and precancerous lesions in varying degrees.MAGE-A1,A5,A6,A8,A9,A10,A11,A12 were highly expressed in OSCC tissues,indicating that they play a certain role in the occurrence and development of OSCC and may be involved in the carcinogenesis mechanism of OSCC.2.The gene expression of MAGE-A increased with the aggravation of oral mucosal epithelial dysplasia(OED)in precancerous lesions.It is suggested that the risk of malignant transformation of precancerous lesions increases with the increase of MAGE-A gene expression in tissues.It is suggested that the abnormal expression of MAGE-A may be involved in the occurrence and development of oral precancerous lesions,and its high expression may be the early event of OSCC.3.MAGE-A1,A5,A6,A8,A9,A10,A11 are highly expressed in clinical III and IV cases,and MAGE-A1,A5,A6,A8,A9,A10,A11,A12 are highly expressed in patients with lymph node metastasis.It is considered that these subtypes may participate in the mechanism of tumor metastasis and invasion of OSCC and play a certain biological role,and may be related to the poor prognosis of OSCC.4.The gene expression of multiple subtypes of MAGE-A is positively correlated.It is inferred that in the progress of OSCC,the subtypes with positive correlation may regulate each other to promote the occurrence and development of OSCC.It is considered that there is a synergistic relationship among members of the MAGE-A family.
