Objective: To improve the "double cannula method" of carbmanda,transfer the liver of Wistar strain rat to SD strain rat,and accurately introduce the combined gene Ctla4-Ig-Ido into the liver of SD rats by plasmid,detect the corresponding biochemical changes and study the mechanism of the formation of immune tolerance in SD rats.Methods: Wistar rats were used as donors and SD rats as receptors.The first group received the combined gene Ctla4-Ig-Ido,the second group received Ctla4-Ig,the third group received Ido,and the fourth group received blank plasmid.One week after the operation,3 rats in each group were randomly selected and sacrificed.Venous blood was collected in the lower chamber,and ALT,AST,TBiL,TNF-?,IL-2,T lymphocyte apoptosis and liver pathological changes in rats were detected.Results: Pre-experimental group early is surgery training.In the experimental group,the indicators of postoperative inflammatory response were significantly increased in the blank group,and no rats survived within 14 days.The first group was lower than the other three groups,and the pathological changes were not significant,and the RAI score was also low.The fourth group had the highest index,the most significant pathological changes and the highest RAI score.Conclusion:The accurate introduction of the interested Ctla4-Ig-Ido gene in rats can reduce the production of various inflammatory markers and alleviate the pathological reaction of liver.Postoperative survival time of rats was also prolonged.Therefore,we concluded that the combined gene Ctla4-Ig-Ido could successfully induce immune tolerance in SD rats.
