[Objective]:To observe allograft rejection in rat orthotopic liver transplantation and intraperitoneal injection of CTLA4-Ig after transplant recipients,to explore the role and mechanism of CTLA4-Ig in producing immune tolerance induced in rat liver transplantation.[Methods]:Building the model of rat orthotopic liver transplantation by the two cuff method,and divided randomly into A.B.C and D groups:wistar to wistar.SD to wistar.SD to wistar treatment group.and wistar to SD.observeing the rejection after rat liver transplantation. Intraperitoneal injection of CTLA4-Ig after the transplantation in contrast to no treatments team,observing the general condition.liver function.the transplanted liver pathological character.and cytokine changes on7and60days after surgery.Throughout.observe the role of CTLA4-Ig in the induction of immune tolerance and liver transplantation.[Results]:①Group A survive longer.and no rejection, group B could survive5-14days after transplantation rejection.group C survive ten to tewenty more time than group B.②Intraperitoneal injection of CTLA4-Ig in SD to wistar group.observed rejection is not obvious after surgery7days.liver biopsy rejection is obviousness. TNF-α,IL-4and IL-10have not significant higher or lower levels.③bservation of SD to wistar injection group on60days after surgery.good general condition.liver biopsy rejection is not serious.cytokines levels of TNF-α,IL-4,IL-10were no appearant changs than normal levels.[Conclusion]:①SD to wistar group has apparently transplant rejection and applicable to observation.②CTLA4-Ig has the effect of anti-liver transplant rejection and inducing immune tolerance of liver transplantation.③The cytokins of TNF-α,IL-4and IL-10can be used as a valuable indicators those reflect the rejection degree.
