Objective: To target the TLR4 / NF-?B inflammatory signaling pathway,focus on the modern common disease of ulcerative colitis(UC),and use Gancaoxiexin decoction to intervene in the acute attack of ulcerative colitis.Research has important practical significance,provides scientific research ideas for UC's traditional Chinese medicine therapy,and provides relevant basis for clinically feasible diagnosis and treatment.Methods: After 1 week of adaptive intervention,Wistar rats were randomly divided into a blank group,a model group,a sulfasalazine group(Western medicine group),six groups of Gancaoxiexin Decoction high,medium and low dose groups,through 2,4-2A nitrochlorobenzene combined with acetic acid compound method was used to establish a rat model of ulcerative colitis.Give Chinese medicine,western medicine,and distilled water by gavage.The blood was collected from the abdominal aorta 2 weeks after the onset.HE staining was used to evaluate the effect of the prescription on the histomorphology of model animals;SP method immunohistochemical method was used to analyze the expression of TLR4 and NF-?Bp65 protein in colonic mucosa tissue;,Whether IL-8,TNF-? and Gancaoxiexin decoction inhibit ulcerative colitis are related to the inhibition of TLR4 / NF-?B signaling pathway and exert their mechanism of action.result:1.Results of renal colon cancer evaluation: There were no significant differences in the mental state,food intake,coat color,etc.of rats in each group before modeling.After the model was built,the intervention rats had a worse mental state than the blank rats,and the food intake and drinking water decreased.The rat hair became sparse,dull,loose stools,and the symptoms of blood in the stool gradually became more serious.After treatment,the rats' mental state gradually recovered and the intake of drinking water increased.,Hair glossiness,loose stool occult blood relief,(all P <0.05),the nominal group weight increase was slightly lower than the blank group(all P <0.05),but significantly above the model group.2.Pathological morphological results of HE staining of colon tissue: The normal group mucosa layer,lamina propria and mucosa muscular layer structure were observed intact under light microscope,no deformation and inflammatory exudation were observed,and there was no necrosis and shedding.Under the microscope,the submucosa showed vasodilation,large tissue edema and diffuse inflammatory cell infiltration;after treatment,the mucosa of the colon tissue of the rat after replacement treatment was clear,and mucosal ulcers or erosions improved to varying degrees.3.Serum ELISA test results: the serum levels of IL-1?,IL-6,IL-8,and TNF-? in the UC model rats were partially blank and significantly increased(all P <0.05),the model group had the highest content,and the remaining groups Both are significantly lower than the model group,and the IL-10 content in the serum of the model group is the lowest,indicating that the body has a resonant response,and the target group is close to or higher than the normal group,resulting in insignificant differences between the groups,indicating that the intervention has a certain therapeutic effect.However,it can not be explained that the appropriate method is used to reduce the advantage;the IL-10 content of the model group is significantly lower than that of the blank group,the middle dose of traditional Chinese medicine,and the high dose(all P <0.01),and the difference between the middle dose group and the remaining groups is statistically significant(All P <0.05).4.Results of tissue immunohistochemistry: Compared with the blank group,TLR4 and NF-?B in the intestinal mucosa of the model group were significantly increased(P <0.05);while compared with the model group,TLR4 in the intestinal mucosa of the other groups of rats The expression of NF-?B was significantly lower than that of the model group(P <0.01);NF-?B and TLR4 had no significant difference between the corresponding groups(P> 0.05).Conclusion: Gancaoxiexin decoction is effective in the treatment of ulcerative colitis rats.Its mechanism may be through inhibiting the activation of TLR4 / NF-?B signaling pathway,thereby reducing the production of pro-inflammatory cytokines and up-regulating the level of anti-inflammatory factors.,Reduce the inflammatory response,thereby improving acute function,and can restore the original structure,therefore,the use of Gancaoxiexin decoction to treat ulcerative colitis can better guide clinical treatment.
