Objective:The purpose of this study was to investigate the effect of hyperbaric oxygen combined with electroacupuncture on myelin related inhibitory proteins(nogo-a and NgR receptors)in rats with spinal cord injury.Methods: 84 female SD rats were randomly divided into 4 groups,followed by model group,hyperbaric oxygen treatment group electroacupuncture treatment group and combined treatment group.Using Allen?s strike method to hit rats,the experimental weight 5 g,height 4 cm..The model group did not intervene after successful modeling,hyperbaric oxygen treatment group was treated with hyperbaric oxygen treatment,treatment pressure was 2.0 ATA,pure oxygen washing tank 10 min,and then decompression 15 min out of the cabin,the total treatment time 85 min,once a day;electroacupuncture treatment group selected rats Dazhui,Shengmen,Yaoyangguan and Changqiang points each into 5 mm dural,using Huatuo brand S.DZ-2 type electroacupuncture therapy instrument,dense wave,2 Hz frequency,moderate intensity,with slight muscle jitter at acupuncture point,suitable for rhythmic contraction of paralyzed muscles of both lower limbs after adaptation,treatment time is 20 min;combined treatment group is combined with hyperbaric oxygen and electroacupuncture treatment.BBB score,HE staining and RT-q PCR were used to detect motor function in rats with spinal cord injury.Results: The BBB scores of the combined treatment group,hyperbaric oxygen treatment group and electroacupuncture treatment group were higher than those of the model group on the 3,7 and 14 th day after injury,which was statistically significant(P < 0.001),and the scores of the combined treatment group were higher than those of the hyperbaric oxygen treatment group and electroacupuncture treatment group(P <0.001),but there was no significant difference between the hyperbaric oxygen group and electroacupuncture treatment group(P > 0.05).In the same group,compared with3,7 and 14 days,the damage of 14 days was 3,7 days BBB score increased significantly,with statistical significance(P < 0.001);HE staining: in the model group,there was bleeding in the background,inflammatory exudation around,partial hemorrhage and necrosis,a large number of inflammatory cells gathered,nuclear shrinkage.After hyperbaric oxygen and electroacupuncture treatment,the glial cells in the injured area increased reactivity,inflammatory cells decreased,apoptosis decreased;3,7 and 14 days after injury,hyperbaric oxygen group and electroacupuncture group The expression of Nogo-A m RNA in the combined group was significantly lower than that in the model group(0.66 ± 0.31,0.72 ± 0.23,0.72 ±0.16),(0.76 ± 0.39,0.80 ± 0.23,0.67 ± 0.41),(0.41 ± 0.31,0.37 ± 0.07,0.40 ± 0.21)(P < 0.05).The expression of Nogo-A m RNA in the combined treatment group was lower than that in the hyperbaric oxygen treatment group and the electroacupuncture treatment group.In addition,there was no significant change in Nogo-A m RNA expression in the same group at different time(P > 0.05).The expression of Ng R m RNA in hyperbaric oxygen group(0.49 ± 0.53,0.52 ± 0.47),electroacupuncture group(0.53 ± 0.58,0.58 ± 0.36)and combined treatment group(0.16 ± 0.13,0.20 ±0.13)were lower than that in model group(0.49 ± 0.53,0.52 ± 0.47),respectively The expression of Ng R m RNA in hyperbaric oxygen treatment group and electroacupuncture treatment group was higher than that in combined treatment group(P < 0.05).There was no significant difference between hyperbaric oxygen group and electroacupuncture group.In addition,the expression of Ng R m RNA in each group did not change significantly(P > 0.05).Conclusion: Both hyperbaric oxygen and electroacupuncture can promote the growth of spinal cord fiber bundle by inhibiting the expression of Nogo-A?Ng R m RNA gene,improve the recovery of nerve function and motor function of rats with spinal cord injury,also reduce the secondary injury after spinal cord injury.Compared with single treatment,combined treatment can better inhibit Nogo-A and Ng R The expression of m RNA gene can promote the growth of spinal nerve fiber and improve the motor function and nerve function of spinal cord injury.
