Objective:To study the chemical constituents of the dried root of Jasminum sambac?L.?Ait,and to explore the active part of root of Jasminum sambac?L?Ait,which plays a sedative role through animal experiments,so as to provide a scientific basis for the further research and development of root of Jasminum sambac?L.?Ait traditional Chinese medicine preparations.Methods:1.The acute toxicity of root of Jasminum sambac?L.?Ait to mice was determined by the determination of half lethal dose(LD500 value),and the sedative effect of root of Jasminum sambac?L.?Ait,extract and different extracts was evaluated by the indicators of total distance,activity speed and activity time of mice,including the detection of spontaneous activity in a dark chamber and the analysis system of spontaneous activity.2.The root of Jasminum sambac?L.?Ait was extracted by the solvent extraction method at room temperature.And 75%ethanol extract was made into ethanol extract aqueous solution and then extracted with petroleum ether,chloroform and water saturated n-butanol.Each layer of extract was separated and purified by chromatographic methods,such as silica gel,octadecyl silane bonded silica?ODS?,hydroxypropyl dextran gel?Sephadex LH-20?,the preparation of thin layer chromatography.The structure of the isolated monomer compound was tested by Nuclear Magnetic spectrum technique,and the structure of the compound was analyzed with the literature data.Results:1.Half of the lethal dose?converted into biogenic dose?of oral root of Jasminum sambac?L.?Ait extract in mice was 125.89 g/kg.The activity distance,activity speed,activity time and other indicators of the mice were detected by the spontaneous activity in the dark chamber.Compared with the control group,root of Jasminum sambac?L.?Ait extract,petroleum ether extract and n-butanol extract have different degrees of difference,showing that they have a sedative pharmacological effect.2.12 compounds were isolated from 75%ethanol extract of root of Jasminum sambac?L.?Ait by room temperature immersion solvent extraction method and various chromatographic chromatography separation methods,and the structures of 12 compounds were identified by spectral method and literature data.3.10 compounds were isolated from the petroleum ether extract of root of Jasminum sambac?L.?Ait extract,and 10 compounds had been structurally identified.The four flavonoids were Boeravinone C?1?,4,11-dihydroxy-9-methoxy-6-ethyoxyl-10-methyl-[1]benzopyrano[3,4-b][1]benzopyran-12?6H?-one?2?,Boeravinone?3?,1?,11-dihydroxy-9-methoxy-6-propoxy-10-methyl-[1]benzopyrano[3,4-b][3]pyrane[5][2]fura-12?6H?-one?4?.The three steroids were Stigmasterol?6?,Daucosterol?7?,7-keto-?5-cholesterol?8?.A triterpenoid was 3-hydroxy-2-ethoxy-24-methyl-enecycloartane?9?.The two other compounds were 4,10-dyhydroxl-16-methyl-13-ethyoxyl-[3]benzopyrano[1]pyran-2-ketone?5?and 5-methyl-2-phenylbenzoxazole?11?.4.2 compounds were isolated from the chloroform extract of root of Jasminum sambac?L.?Ait extract,and the structures of two compounds were identified as one flavonoid:Boeravinone H?10?and one other compound,6-methyl-1,3,8-trihydroxyanthraquinone?12?.Conclusion:1.The pharmacological effects of root of Jasminum sambac?L.?Ait on central inhibition and sedation and sleep were verified,and different solvent extracts of alcohol extract showed differences in pharmacological activity intensity,which had certain guiding significance for the study of active sites.2.There were 12 compounds were isolated from root of Jasminum sambac?L.?Ait,and 12 structure compounds were identified,including two compounds 4,5 for new compounds.A compound 3 was isolated from this genus for the first time,and four compounds 1,2,11,12 were isolated from this plant for the first time,which laid a foundation for clarifying the chemical composition of root of Jasminum sambac?L.?Ait.
