Objective:To study the effect of total flavonoids of Jasminum nudiflorum Lindl on chronic non-bacterial prostatitis?CNP?model animals and to study its mechanism of action on CNP.Methods:CNP model mice were replicated by removing bilateral testes and injecting estradiol benzoate.One week after the testicles were removed,estradiol benzoate was subcutaneously injected for 30 days and administered intragastrically for 30 days.0n the10th,20th and 30th days,the body weight of each mouse was measured.On the 7th,14th,21th and 28th day,the daily drinking water and daily urine output of each group of mice were measured.After 2 hours of administration on the 30th day,the on-site activity index was measured.On the morning of the next day 30 days after the administration,blood was taken from the eyeball.The mice were killed by cervical dislocation,and the prostate and kidney were removed.The weights of the prostate and kidney were weighed and the wet weight index was calculated.Divide the prostate into two parts,the first one was used to observe the number of white blood cells and the density of lecithin bodies,the second part was used to detect the levels of IL-6,IL-8,IgA,IgG,IFN-g,and TGF-bin the prostate homogenate.The stratified and centrifuged mouse serum was used to measure the levels of TNF-aand IL-1b.The CNP model rats were duplicated by removing the bilateral testes and injecting estradiol benzoate.One week after the testicles were removed,estradiol benzoate was subcutaneously injected for 30 days and administered intragastrically for 30 days.Each rat was weighed before administration on the 10th,20th,and 30th day,and drinking water was measured on each of the rats on the 7th,14th,21th,and 28th day after the administration.The amount and the daily urine output were performed on the 29th day after the administration of cystostomy,and 24 hours after the last administration,the urodynamic parameters of the rats were measured.After the completion of the bladder pressure measurement system,the rats were anesthetized and blood was collected using the abdominal aorta.The rats were sacrificed and the prostate,epididymis,bladder and kidney were removed.The weights of the prostate and kidney were weighed and the wet weight index was calculated.The prostate was cut into three parts.The first one was placed in liquid nitrogen and the levels of P-P38MAPK were detected by Western blot,and the level of P38 mRNA was detected by Q-PCR.The second homogenate was used to detect the levels of IL-2,IL-10,COX-2,MIP-1a,MMP-9,P38MAPK,and P-P38MAPK.The third part was placed in a 4%solution of formaldehyde with epididymis,bladder,and kidneys.The changes in the morphology of the prostate,bladder,kidney,and epididymis were observed.The stratified and centrifuged serum was used to determine the levels of TNF-aand IL-1b.The CNP model rats were duplicated by means of bilateral prostatitis injections of 25%of adiponectin.One week after surgery,the rats were continuously administered intragastrically for 21 days.Each rat was weighed before administration on the 10th and20th days.Daily drinking water intake and daily urine output were measured on the 7th,14th,and 20th day after administration,and the on-field activity index was measured on the21st day after 2 hours of administration.After 24 hours of the last administration,the rats were anesthetized and blood was collected using the abdominal aorta.The rats were sacrificed and the prostate,testis,epididymis,bladder,kidney,and liver were removed,and the weights of the prostate,testis,and epididymis were weighed,and the wet weight index was calculated.Divide the prostate into three parts.The first one was used to observe the number of white blood cells and the density of lecithin bodies;The second part was used to test the content of PGE2,BK,LTB4,SOD and MDA in the homogenate;The third portion was stored in 4%formaldehyde with testis,epididymis,bladder,kidney,and liver,and the change in tissue morphology was observed.The serum after centrifugation was used to measure the levels of TNF-a,IL-1b,T,and ACP.Plasma was used to determine the levels of TXB2 and 6-keto-PGF1a.Results:By observing the total flavonoids of Jasminum nudiflorum Lindl primrose extract on the testis-evoked estrogen-induced CNP model mouse.The mice in the model control group had little change in body weight gain,and the amount of urine output and water intake decreased,especially around the 14th day of modeling.In the opening event,the time for exercise,the duration of stay in the surrounding area,the number of legs lifted,and the total number of spanning squares have increased.The levels of TNF-aand IL-1bin serum increased significantly;the number of leukocytes in prostate tissue increased significantly,and the lecithin body density score decreased significantly;The contents of IL-6,IL-8,IgA,IgG,IFN-g,and TGF-bin the prostate tissue homogenate were significantly increased.The body weight,urine output,and water intake of total flavonoids in the treatment group were higher than those in the model control group.The indicators in the opening activities were relatively reduced,the number of white blood cells was decreased,the density of lecithin bodies was increased,and various factors in serum and prostate tissues were effectively improved,and the lesions in the prostate,epididymis,bladder,and kidney were improved.By observing the total flavonoids of Jasminum nudiflorum Lindl primrose extract on the testis-evoked estrogen-induced CNP model rats,we found that the weight of rats in the model control group was significantly lower than that of the administration group,the urine output and the amount of drinking water were decreased,the urinary dynamic index of the urination threshold was increased,the urination interval was shortened,and the maximum urination pressure was decreased,serum TNF-aand IL-1blevels increased significantly.The contents of COX-2,IL-2,MMP-9,P38MAPK,P-P38MAPK and MIP-1ain the homogenate of prostate tissue were significantly increased,the content of IL-10 was significantly decreased,the expression of P38 mRNA was increased,and the P-P38MAPK proteins were increased.Increased expression levels.The organ index of the prostate and kidneys was reduced,and the corresponding pathological changes in the prostate,epididymis,bladder,and kidney appeared.The body weight,urine output,and water intake of rats treated with total flavonoids from Jasminum nudiflorum Lindl were higher than those of the model control group.The indicators in urodynamics were relatively reduced,and various factors in serum and prostate tissues were effectively improved,and the lesions in the prostate,epididymis,bladder,and kidney were alleviated.By observing the total flavonoids of winter jasmine flowers on the CNP model rats induced by Xiaozhiling,it was found that the weight of rats in the model control group was significantly lower than that of the administration group,and the urine output and drinking water were decreased,and the opening time was measured in the exercise time and peripherals.Stay time,number of legs lifted,total number of spans increased.Serum levels of TNF-a,IL-1b,and ACP increased significantly,T significantly decreased,plasma TXB2increased significantly,6-keto-PGF1adecreased significantly,and MDA,PGE2,LTB4,and BK levels in prostate homogenates increased significantly.The SOD content was significantly reduced,the number of white blood cells in the prostate tissue increased significantly,the lecithin body density score significantly decreased,and the prostate,testis and epididymal organ index increased.Prostate,testis,epididymis,bladder,kidneys,and liver have corresponding pathological changes.The body weight,urine output,and water intake of rats treated with total flavonoids in the Jasminum nudiflorum Lindl primal flavone were higher than those in the model control group.The indicators of opening activities were relatively lightened,and various factors in the serum and prostate tissues were effectively improved.The prostate,testis,epididymis,and bladder were significantly improved,kidney,liver lesions have a mitigating effect.Conclusion:Total Flavonoids of Jasminum nudiflorum Lindl can improve the levels of anti-inflammatory and pro-inflammatory factors in CNP model animals,enhance the antioxidant capacity of model animals,relieve blood circulation disorders and pain responses,reduce the anxiety behavior of model animals,and relieve urinary disorders,improve systemic immunity.At the same time,the regulation of P38MAPK signaling pathway has been improved,indicating that total flavonoids of Jasminum nudiflorum Lindl can effectively relieve inflammation symptoms and treat CNP.
