Objective:To study the spectrum-effect relationship of the anti-oxidation and bacteriostasis of Rose roxburghii,combining HPLC fingerprints of Rose roxburghii polar parts extracts and data of anti-oxidation and antibacterial efficacy,partial least squares method and grey correlation method were used to obtain the spectrum-effect relationship of antioxidant and antibacterial effects of Rose roxburghii,to investigate the correlation between the common peaks of HPLC fingerprints of different polar parts of Rose roxburghii on the antioxidant and bacteriostatic effects of Rose roxburghii,confirm the contribution of each component peak to the efficacy,preliminary clarification of the material basis of anti-oxidation and bacteriostasis of Rose roxburghii different polar parts.Methods:By investigating the chromatographic conditions of HPLC,the fingerprints of Rose roxburghii different polar parts were determined,DPPH method and ABTS method were used to study the antioxidant activity of Rose roxburghii different polar parts;to study the antibacterial effect of Rose roxburghii different polar parts against five types of bacteria,Staphylococcus aureus,Bacillus subtilis,Enterococcus faecalis,Proteus vulgaris,and Pseudomonas aeruginosa by bacteriostatic circle method,using partial least squares method and grey correlation degree to correlate the"spectrum"and"effect"of Rose roxburghii;a spectrum-effect relationship model of anti-oxidation and bacteriostasis of Rose roxburghii was constructed;to study the contribution of the common peak of Rose roxburghii different polar parts to the antioxidant and bacteriostatic effects.Results:The fingerprint map of Rose roxburghii five different polar partswas established,20common peaks were calibrated and 5 common peaks were identified,peak 1 is ascorbic acid,peak 5 is gallic acid,peak 7 is protocatechuic acid,peak 16 is ellagic acid,and peak 17 is rutin;the scavenging effect of the 5 kinds of polar parts samples on DPPH free radicals is ethyl acetate part>n-butanol part>water part>methanol part>ethanol part,the ethyl acetate site has the best scavenging effect on DPPH free radicals(EC50=2.46),It is equivalent to the clearance effect of the reference substance Vc(EC50=2.26);the effect of the five polar parts samples on the removal of ABTS free radicals is ethyl acetate part>n-butanol part>methanol part>ethanol part>water part,The ethyl acetate site has the best scavenging effect on DPPH free radicals(EC50=23.33?g/ml);Rose roxburghii five polar parts samples have an inhibitory effect on Staphylococcus aureus,Bacillus subtilis and Proteus vulgaris,the methanol,ethanol,n-butanol and ethyl acetate of Rose roxburghii have an inhibitory effect on Enterococcus faecalis,the water,methanol,ethanol and ethyl acetate parts of Rose roxburghii have an inhibitory effect on Pseudomonas aeruginosa?bacteriostatic circle?7mm?;Gray correlation results show Rose roxburghii five polar parts samples achieve antioxidant and bacteriostatic effects through it's internal"effective component group",several common peaks with greater influence rank DPPH free radical scavenging rate as follows:16>10>17>8>9>13>4,The correlation degree of ABTS free radical scavenging rate is 15>8>12>13,The order of relevance of inhibiting Staphylococcus aureus is:3>13>18>17,The relevance of the inhibition of Bacillus subtilis ranks as 16>10>15>8,The relevance ranking of the inhibition of Enterococcus faecalis was 10>16>8>9>17,The relevance of the inhibition of common Proteobacteria ranks as 6>16>9>13>15,The correlation degree of the inhibition of Pseudomonas aeruginosa was 13>3>17>18>4;partial least squares results indicate that the contribution of the common peak of Rose roxburghii to DPPH free radical scavenging rate is 11>4>8>1>19>7>5>13>12>15>17>13>18>9>10,the order of contribution to ABTS free radical scavenging rate is 11>15>19>7>12>5>1>8>4>10>13>3>17>2>20>16,the order of contribution to the inhibition of Staphylococcus aureus was 18>4>8>14>11>2>9>10>12>15>19>1>7,the order of contribution to the inhibition of Bacillus subtilis is“16>20>13>3>17>5>6>12>15>1>7>19>10>2”,The contribution to the inhibition of Enterococcus faecalis ranks as16>20>2>14>10,the order of contribution to the inhibition of common Proteobacteria was 18>6>9>17>4,the order of contribution to the inhibition of Pseudomonas aeruginosa was 16>6>9>17>4>18>14>12>1>20>13>10>19>15>7>5>8.Conclusion:Rose roxburghii five polar parts samples fingerprint can be used for quality control and evaluation of Rose roxburghii,the scavenging effect of Rose roxburghii on DPPH free radical?ABTS free radical and Inhibition on five kinds of bacteria is the result of the combined action of multiple ingredients,through the spectrum-effect relationship,it is possible to determine the"effective component group"of the effect of traditional Chinese medicine,provide new methods for the quality evaluation and pharmacodynamic research of traditional Chinese medicine.
