Chapter 1:The Expression of GLUT3 in Pituitary Adenomas and Its Relationship with Ischemia and HypoxiaObjective To investigate the role of expression of glucose transporter 3(GLUT3)and glucose transporter 1(GLUT1)in pituitary adenomas,including effects on tumor ischemia,hypoxia and cystic changes.Methods A total of 203 surgically resected PA tumor samples that were pathologically confirmed were collected from January 2013 to April 2017 at the Department of Neurosurgery in this hospital.Complete medical records,images,and pathological specimens were obtained.To observe expressions of GLUT3 and GLUT1 in tissue of pituitary adenoma by immunohistochemistry,and to analyze the relationship between the age,Knosp score,Ki-67,cystic change,hormone type,and tumor size in the groups.Results The expression rate of GLUT3 in cystic group of pituitary adenomas was 54.5%(72/132),and the expression rate in non-cystic change group was 36.6%(26/71).The cystic group was higher than the non-cystic change group(P = 0.018).Proportions of GLUT3-positive staining of microadenomas,macroadenomas,and giant adenomas were 22.7%(5/22),50.4%(66/131)and 54.0%(27/50),respectively(P = 0.022).In cases of prolactin adenoma,GLUT3-positive staining was predominant in cell membranes(P <0.001),while in cases of follicle-stimulating hormone or luteotropic hormone adenoma,we found mainly paranuclear dot-like GLUT3 staining(P = 0.025).In other hormonal adenomas,GLUT3 was only partially expressed,and the intensity of cell membrane or paranuclear punctate staining was weak.In contrast to GLUT3,GLUT1 expression was not associated with pituitary adenomas.Conclusion Our results indicate that the expression of GLUT3 in pituitary adenomas is closely related to cystic change and hormonal type.This study is the first to report a unique paranuclear dot-like GLUT3 staining pattern in pituitary adenomas.Chapter 2:The Study on Expression of GLUT3 and HIF-1? in MeningiomasObjective To investigate the role of expression of glucose transporter 3(GLUT3)and hypoxia inducible factor-1? protein(HIF-1?)in meningiomas,to analyze the correlation of expression of GLUT3 and HIF-1? with the imaging features,pathological grade of meningiomas.Methods A total of 167 surgically resected meningiomas samples that were pathologically confirmed were collected from January 2015 to December 2017 at the Department of Neurosurgery in this hospital.Complete medical records,images,and pathological specimens were obtained.The postoperative tumor specimens were pathologically graded according to the WHO 2007 classification criteria for central nervous system tumors,divided into low grade meningiomas(estimated as WHOI)and high grade meningiomas(estimated as WHOII or WHOIII).Peripheral edema assessment was performed on magnetic resonance images(MRI),and the degree of peritumoral edema(PTBE)of meningiomas was graded using the Steinhoff classification method.To observe expressions of GLUT3 and HIF-1? in tissue of pituitary adenoma by immunohistochemistry.Results Proportions of GLUT3-positive staining of high grade group and low grade group were71.4%(15/21)and 30.8%(45/146),respectively(P<0.001).Proportions of HIF-1?-positive staining of high grade group and low grade group were 76.2%(16/21)and 44.5%(65/146),respectively(P = 0.007).GLUT3-positive expression in meningioma PTBE 0,I,II,and III were 12.2%(10/82),35.5%(11/31),62.1%(18/29),84.0%(21/25),respectively(Bonferroni-corrected,P < 0.001,?/6=0.008).HIF-1?-positive expression in meningioma PTBE 0,I,II and III were 35.4%(29/82),51.6%(16/31),58.6%(17/29),84.0%(19/25),respectively(Bonferroni-corrected,P=0.002,?/6=0.008).Spearman correlation analysis was used to analyze the correlation There was a correlation between the expression of GLUT3 and HIF-1? in meningiomas(r=0.330,P <0.001).Conclusions This study found for the first time that GLUT3 has a unique map-like staining pattern in meningiomas.GLUT3 may play a key role in the process of peritumoral edema induced by ischemia and hypoxia.GLUT3 and HIF-1? may be involved in the formation of peritumoral edema of meningiomas,and the phenotype with PTBE in meningiomas may be more aggressive.By comparing the tumor cell proliferation ability with the MRI signs,the biological characteristics are analyzed,which provides reliable information for future clinical treatment and prognosis evaluation.
