Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection has the effects of dilating blood vessels,resisting platelet aggregation,improving microcirculation,and improving myocardial ischemia tolerance.It is often used in the treatment of occlusive cardiovascular and cerebrovascular diseases.In order to further clarify its chemical composition,screen its new pharmacological activity,and expand its clinical application,this paper uses UPLC-Q / TOF-MS and other separation technologies,UNIFI natural product analysis platform,compound standard comparison and other identification methods.Multivariate statistical analysis conducted in-depth research on the chemicals of this injection;meanwhile,a rat model of acute blood stasis induced by ice water bath plus epinephrine was constructed,and the mechanism of anti-blood stasis activity of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection Preliminary research has achieved the following results:1.Study on the full components of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection based on UPLC-Q / TOF-MS technologyFor the first time,UPLC-Q / TOF-MS technology combined with the UNIFI natural product analysis platform was used to analyze the full composition of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection.As a result,a total of 40 chemical components such as flavonoids,esters,quinones and terpenes were identified,providing detailed information on the chemical composition of the injection.2.Study on the brain composition of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection based on UPLC-Q / TOF-MS technologyFor the first time,the UPLC-Q / TOF-MS technology combined with multivariate statistical analysis was used to study the brain-entering components of alviae Miltiorrhizae and Ligustrazine Hydrochloride injection.A total of 7 chemical constituents,such as danshensu,ligustrazine,ethyl protocatechuate,monomethyl shikonate,rosmarinic acid,ligandolide Q,and linoleyl alcohol,were identified in the brain tissues administered.The injection provides a reference for the substance basis in the brain.3.Metabolites of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection Based on UPLC-Q/TOF-MS TechnologyUPLC-Q / TOF-MS technology and multivariate statistical analysis were used for the first time to study the metabolites of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection in rats.A total of 15 metabolites were found in the plasma,urine,feces,and bile of the administered rats,of which 2 prototype metabolites were danshensu and ligustrazine,10 metabolites of danshensu,and metabolites of ligustrazine 3 These involve multiple metabolic pathways such as hydroxylation,carboxylation,sulfation and glucuronidation.4.Preliminary study on the anti-blood pharmacological activity of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injectionA preliminary study on the anti-blood stasis activity of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection was carried out for the first time.As a result,compared with the model group,the administration group could reduce the whole blood and plasma viscosity of the model group,and significantly reduce the GMP-140 and vWF content in the plasma.The above results show that Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection can treat acute blood stasis caused by ice water bath plus adrenaline,can improve blood rheology parameters,reduce the content of GMP-140 protein and vWF factor,and has anti-blood stasis effect.5.Prediction of anti-blood stasis mechanism of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection based on network pharmacology research methodUsing the network pharmacology research method and combining the previous research results of chemical constituents of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection to predict the anti-blood stasis mechanism of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection.As a result,through database analysis,31 active ingredients,38 targets were predicted,and 10 pathways were related to blood stasis diseases.The mechanism of synergistic treatment of blood stasis diseases by this injection was initially revealed.6.Study on anti-blood stasis mechanism of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection based on metabolomicsUsing metabolomics technology to explore the anti-blood stasis mechanism of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection at the overall level,the results showed that Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection significantly reversed the abnormal changes of 22 endogenous metabolites caused by acute blood stasis.Regulation of sphingolipid metabolism,steroid hormone biosynthesis,retinol metabolism,arachidonic acid metabolism,α-linolenic acid metabolism,linoleic acid metabolism,glycerophospholipid metabolism,etc.play an anti-blood stasis effect.To sum up,this thesis enriches the chemical constituents of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection,identifies the brain-entering and metabolic components of the injection,screens the pharmacological activity of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection against blood stasis,and adopts network pharmacology And metabolomics technology to predict and study the mechanism of anti-blood stasis effect of injection.The chemical composition and pharmacological activity of Salviae Miltiorrhizae and Ligustrazine Hydrochloride injection were systematically studied in the paper,which provided a reference for further development of salvia ligustrazine injection.
