Paeonia Lactiflora and Radix Paeoniae Rubra are both dry roots of Ranunculaceae as plant medicinal materials.Paeonia Lactiflora is mainly produced in Anhui,Zhejiang and other places;Radix Paeoniae Rubra is mainly produced in Inner Mongolia and Liaoning.As auxiliary medicinal material,these two have both the same points and different points in pharmacology.Due to the different origins,batches,and production techniques of the medicinal materials,the effective ingredient content is also different,and the difference in the efficacy is more and more significant.With the advent and widespread application of new formulations such as formulated granules,the quality control of formulations has become the focus of attention today.As the predecessor of granule formulations,the study of quality standards is particularly important.In this project,white peony and red peony are prepared into formula granules,and the quality evaluation of Paeonia Lactiflora and Radix Paeoniae Rubra standard decoction and Paeonia Lactiflora and Radix Paeoniae Rubra formula granules are carried out,with a view to standardizing the clinical application of Paeonia Lactiflora and Radix Paeoniae Rubra formula granules.Research content of this topic:1.Study on the quality standard of Paeonia Lactiflora and Radix Paeoniae RubraIn this experiment,the extraction conditions of lyophilized samples of Paeonia Lactiflora and Radix Paeoniae Rubra were investigated by high performance liquid chromatography,and the water solvent was determined as the best solvent for extracting the lyophilized samples of Paeonia Lactiflora and Radix Paeoniae Rubra decoction.The optimal chromatographic conditions were determined as : Gradient elution with acetonitrile-0.1% formic acid aqueous solution,column temperature25 ?,flow rate 0.4ml / min,detection wavelength is 230 nm.UPLC-Orbitrap technology was used to identify the main four chemical components of Paeonia Lactiflora: galloyl dextrose,paeonia lactiflora ester glycosides,paeoniflorin and benzoyl paeoniflorin.Nine characteristic peaks of Paeonia Lactiflora and seven peaks of Radix Paeoniae Rubra were determined by standard decoction method The characteristic peaks with good degree of stability and good stability.Based on 9characteristic peaks of Paeonia Lactiflora and 7 characteristic peaks of Radix Paeoniae Rubra,the similarity of 15 batches of Paeonia Lactiflora and Radix Paeoniae Rubra characteristic map were compared respectively.Other data explore the similarities and differences of Paeonia Lactiflora and Radix Paeoniae Rubra decoctions from different sources and different origins.It provides a basis for the subsequent determination of multi-component quality control,content index and content limit of Paeonia Lactiflora and Radix Paeoniae Rubra formula particles.2.Preparation of Paeonia Lactiflora and Radix Paeoniae Rubra formula granules and their quality standardsThe Paeonia Lactiflora and Radix Paeoniae Rubra are concentrated and evaporated to dryness according to the decoction extraction method.After grinding and crushing,by evaluating the softness and hardness of the granular soft materials,dextrin is used as the first choice auxiliary material,and the material ratio is 1: 1.4.After the dosage form is made,the pharmacopoeia granule regulations are taken as the standard to confirm that the granules meet the quality requirements.Subsequent HPLC method was used to investigate the content of Paeonia Lactiflora and Radix Paeoniae Rubra granules(including specificity,linearity,precision,reproducibility,sample recovery rate,stability).The results showed that the method was reliable,accurate,strong specificity and good reproducibility.The content of each index component of fifteen batches of white Paeonia Lactiflora and Radix Paeoniae Rubra standard decoction,the content of each index component of fifteen batches of Paeonia Lactiflora and Radix Paeoniae Rubra formula granules were measured and the transfer rate of each component was calculated.The transmission rate of Paeonia Lactiflora Decoction-Formula granules: paeonilactone is 30-100%,paeoniflorin is 68-98%,benzoyl paeoniflorin is 27-67%.The control substance that can be stably delivered in the process of Paeonia Lactiflora decoction-particles is paeoniflorin.The transmission rate of Radix Paeoniae Rubra Decoction-Formula granules: catechin delivery rate is78%-99%;paeoniflorin is 78-99%;paeoniflorin is 89-99%;benzoyl paeoniflorin is70-102%.It can be concluded from this that the components that can be stably delivered in Radix Paeoniae Rubra decoction-particles are catechins,paeoniflorin,and paeoniflorin.It provides a certain reference for the content determination and quality standard establishment of Paeonia Lactiflora and Radix Paeoniae Rubra standard decoctions and formula granules.3.A preliminary study on the hypoglycemic effect of Paeonia lactiflora extractAlloxan selectively destroys ?-cells of mouse islets,reduces insulin secretion and enters the blood circulation,which in turn leads to increased blood glucose in mice.It is commonly used in diabetic mice in animal experiments.High concentration of insulin induces HepG2 cells to be used as an insulin resistance model.These two models are commonly used to explore the mechanism of action of drugs on diabetes.The results are stable and good,and the confidence interval is large.Diabetic mice with successful modeling of alloxan have a high probability of having obvious signs and symptoms of diabetes such as polydipsia,polyphagia,polyuria,weight loss,and blood glucose levels above 15 mmol / L.After 30 days of intragastric administration of the extract of Paeonia lactiflora,the above-mentioned diabetes signs in the high-medium-low-dose group improved significantly.After the experimental indicators of diabetes were measured,each treatment group had a good hypoglycemic effect.The high-dose white peony group has a significant effect in reducing fasting blood glucose,improving glucose tolerance,increasing muscle glycogen content,and promoting insulin secretion.,But for increasing serum insulin content,the effect of liver muscle glycogen content is not significant.In the insulin resistance experiment,the extracts of paeonia lactiflora and paeoniflorin can promote the absorption of peripheral glucose by liver cells and improve insulin resistance.The effect of paeoniflorin group and white peony extract group on insulin improvement is similar,and the content of paeoniflorin in white peony extract is about 2.5%,and the content of paeoniflorin in white peony extract is less than 0.1mg / ml,which means In addition to the effect of paeoniflorin on improving insulin resistance,there are still other substances that can assist in improving insulin resistance.
