Objective:To study the mechanism of Sanguisorba tannin elevation in bone marrow suppression of leukocytes in mice by metabonomics.Method:SPF grade KM mice were randomly divided into normal group,model group and sanguisorba tannin group of 10 mice in each group.The model group and the sanguisorba tannins group were intraperitoneally injected with cyclophosphamide100mg·kg-11 to induce the mice to establish a bone marrow inhibition model.On the first day of the experiment,the tannin group was ig tannin aqueous solution at the dose of 10mg·kg-1,and the model group was gavage to purified water at the same dose for 5 consecutive days.On the 6th day,blood was taken from orbit of mice of each experimental group,and bone marrow and hematopoietic stem cells were separated.The number of white blood cells,DNA content of bone marrow cells and hematopoietic stem cells in peripheral blood of mice of each experimental group were detected,and the changes of serum metabolism profile of mice of each experimental group were analyzed by LC-MS.Results:Compared with the model group,the number of white blood cells in the tannin group was significantly increased?P<0.05?,the content of DNA in bone marrow cells was significantly increased?P<0.05?,and the number of hematopoietic stem cells was significantly increased?P<0.05?.By serum metabolic profile analysis,from the model group and normal group were selected 35 different metabolites,model group and sanguisorba tannins,a total of 30 kinds of differences metabolites,screen out the VIP>1.0,the fold-change acuity 1.5 or 0.667 or less and p-the intersection of the value of 0.05 or less,get a cytarabine,D-glucose-6 phosphate,alpha ketone sugar isovaleric acid,lauric acid variables,four differences compared with model group,sanguisorba tannins mice serum cytidine china-arab sugar,D-glucose-6phosphate,alpha ketone sugar isovaleric acid,lauric acid have significant potential metabolic marker callback.Six related metabolic pathways were involved:pentose phosphate pathway,starch and sucrose metabolism,isoleucine and valine leucine metabolism,galactose metabolism,glycolysis and gluconeogenesis,amino acid and nucleotide metabolism.Conclusion:The increase of Sanguisorba tannins-mediate dleukocytes on myelosuppressive in mice may be related to the regulation of pentose phosphate pathway,amino acid and nucleotide metabolism.
