The global morbidity and mortality of cancer are growing rapidly.It is estimated that there will be 18.1 million new cancer cases and 9.6 million cancer deaths in the world in 2018.The new cases and death in China account for about 25%of the new cases and death in the world.It is an effective way to search for an effective and low-toxicity anti-tumor drug in the Chinese traditional medicine which has been proved for a long time.In this paper,two homogenous polysaccharides were purified from chaenomeles speciosa(Sweet)Nakai,and their structures were identified.The homogenous polysaccharide CSP-W-2,CSP-0.2-1,they were separated by DEAE-Sepharose Fast Flow and Superdex 200,CSP-W-2 had a molecular weight of 8.7×10~3Da,and the monosaccharide was composed of glucose,galactose,mannose,arabinose,xylose and rhamnose,3.7:3.2:0.9:1.7:0.4:0.02.The molecular weight of CSP-0.2-1 was 7.3×10~4Da,and the sugar composition was rhamnose,arabinose,xylose,mannose,glucose,galactose 4.0:1.5:9.3:0.5:1.0:3.4.The results of monosaccharide composition need to be confirmed because CSP-0.2-1 contained uronic acids.CCK-8 assay was used to detect the inhibition effect of two homogenous polysaccharides on Hep G2 cells.It was found that the CSP-W-2 isolated from the water had a better inhibitory effect on Hep G2.The Hep G2 cells were treated with 400?g/ml,200?g/ml,100?g/ml and 50?g/ml CSP-W-2,the inhibitory was found to be quantitatively dependent on the concentration.Through inverted microscope observation,it was found that csp-w-2 could inhibit Hep G2 proliferation.Through Hoechest33258 staining,it was found that Hep G2 cell nucleus fluorescence increased significantly after CSP-W-2 treatment,and cell nucleus shrinkage and apoptosis.Through methylation analysis,CSP-W-2 was found to be connected in various ways,including Terminal-Ara,T-Xyl,1,3-Ara,1,5-Ara,T-Man,T-Glc,1,4-Glc,1,4-Gal,1,6-Glc,1,4,6-Man,1,4,6-Gal,1,4,6-Glc.After partial acid hydrolysis,and arabinose was basically hydrolyzed completely and partially hydrolyzed with glucose,indi cating that its branched chain was composed of arabinose and glucose,and the main chain was composed of galactose and glucose and a small amount of mannose.
