Objective:This thesis,based on the models of aging mice movement,explores the effect of resistance training and Sirtl on skeletal muscle mitochondria metabolism and related enzyme of aging mice to illustrate the mechanism of mitochondrion metabolism via Sirtl activation.Methods:Establishing a model of 3-month male mice which were injected D-galactosamine(500mg/kg.day)at the nape.The 36 male C57BL/6 mice randomly assigned to aged control group(OC),aged exercise group(OE),aged with EX527 treatment group(OX),aged with resveratrol treatment group(OR),aged exercise with EX527 treatment group(OEX)and aged exercise with resveratrol treatment group(OER).The mice of OE,OEX and OER groups were supplemented with the exercise of laddering by loading for 4 weeks.When the training finished,the resveratrol were injected into the mice of OR and OER groups through subcutaneous injection at the nape,and the mice of OX and OEX groups were injected with EX527 at the same areas,measuring the total protein content,succinate dehydrogenase(SDH),cytochrome C and ATPase activity of the rectus femoris tissues after 4 weeks.Results:(1)Compared with the OC group,the OR group's enzyme activity of SDH significantly increased(p<0.05);the OER group's SDH activity increased largely(p<0.01).Multi-factor analysis showed that resistance exercise resulted in significant increase in SDH(p<0.01);and the effect of RV and EX527 on SDH also significantly increased(p<0.01).(2)Compared with the OC group,the OE group's ATPase activity significantly increased(p<0.05);the OR group's ATPase activity significantly increased(p<0.05);the OER group's ATPase activity increased largely(p<0.01).Multi-factor analysis showed that resistance exercise resulted in significant increase in ATPase activity p<0.05),and the effect of RV and EX527 on ATPase activity also significantly increased(p<0.01).(3)Compared with OC group,the OE group cytochrome C significantly increased(p<0.01);the OX group's cytochrome C significantly decreased(p<0.05);the OR group's cytochrome C significantly increased(p<0.05);the OER group's cytochrome C increased obviously(p<0.01).Multi-factor analysis showed that resistance exercise resulted in significant increase in cytochrome C(p<0.01),and the effect of RV and EX527 on cytochrome C also significantly increased(p<0.01).Conclusion:(1)Four-week resistance training and Sirtl can promote the tricarboxylic acid cycle in mitochondria,improving the ability of aerobic metabolism of aging skeletal muscle mitochondria.(2)Four-week resistance training and Sirtl can enhance the enzymatic activity of ATPase,and improve the fimction of aging mitochondria.(2)Four-week resistance training and Sirtl can increase the content of cytochrome C,improving the ability of respiratory chain of aging skeletal muscle mitochondria.
