Object:(1)To investigate the inhibitory effect and interaction of aerobic exercise and Oyster Peptide(OP)supplementation on lung cancer growth in tumor-bearing mice.(2)Analyzing the changes of cellular immune function and MicroRNAs in tumor tissues.To explore the possible reasons for aerobic exercise and OP supplementation inhibit the growth of lung cancer in tumor-bearing mice.Methods:Select 48 clean male mice(C57BL/6J),culture LL/2 cell line of mice.The cell concentration was adjusted to 6×106/ml and inoculated subcutaneously into the armpit of the right forelimb of mice,each of which was 0.2 ml.The whole process was strictly aseptic and completed within 40 minutes,and the Lewis lung cancer model of mice was established.They were randomly divided into four groups:tumor-bearing control group(C),tumor-bearing oyster peptide intervention group(OP),tumor-bearing aerobic exercise intervention group(E)and tumor-bearing oyster peptide plus aerobic exercise intervention group(OPE),12 mice in each group.Group C does not exercise(Feed according to the standard diet);The group E was swimming without weight for 60mins 6 time each week;The group OP was drenched OP 30 minutes after each training(supplementary dose of 1.8g/kg),training activity be held six times per week;Weekly measurements of body weight and tumor volume in tumor-bearing mice.Sampling after 5 weeks,the weight of tumors,the percentage of CD3+,CD4+,CD8+T cells,NK cells and CD4+CD25+CD127-Treg cells in blood,the apoptotic rate,necrosis rate of tumor tissue and the expression of microRNA-21 and microRNA-218 were measured,At the same time,observation the number of nodules on the surface of the lung and the structure of tumor tissue.Results:(1)The tumor inhibition rate of subcutaneous tumor mice in each group was 26%in the OP group,16%in the E group,and 24%in the OPE group.The metastasis inhibition rate of tumor-bearing mice in each group was 60%in the OP group,46%in the E group,and 54%in the OPE group.Two-way ANOVA analysis of C,OP,E and OPE showed that there was no significant interaction between aerobic exercise combined with OP supplementation on the weight of tumors and the number of pulmonary nodules(P>0.05);OP supplementation extremely significant reduced the weight of tumors(P<0.01)and the number of pulmonary nodules(P<0.05);aerobic exercise reduced the weight of tumors and the number of pulmonary nodules,but the difference was not significant(P>0.05).(2)Two-way ANOVA analysis of C,OP,E and OPE showed that there was no significant interaction between aerobic exercise and OP supplementation on the increase of necrosis rate and apoptosis rate of lung cancer cell(P>0.05);OP supplementation significantly help increased the necrosis rate and apoptosis rate of tumor tissue(P<0.05);aerobic exercise increased the necrosis rate of lung cancer cell,but the difference was not significant(P>0.05).Oxygen exercise can extremely significant increase the apoptotic rate of lung cancer cell(P<0.01).(3)Two-way ANOVA analysis of C,OP,E and OPE showed that Aerobic exercise combined with OP supplementation had no significant interaction with the increase of CD3+?CD4+?CD8+T cells,NK cells and CD4+/CD8+ratio in blood of tumor-bearing mice(P>0.05).OP supplementation could significantly increase the number of CD3+?CD8+T cells,NK cells and CD4+/CD8+ratio in blood of tumor-bearing mice(P<0.05)and the number of CD4+T cells extremely significant increase(P<0.01).Aerobic exercise could increase the number of CD3+?CD4+?CD8+T cells,NK cells and CD4+/CD8+ratio in blood of tumor-bearing mice,but not significantly(P>0.05).There was no significant interaction between aerobic exercise and OP supplementation on the decrease of CD4+CD25+CD127-Treg cells in the blood of tumor-bearing mice(P>0.05).OP supplementation could extremely significant decrease the number of CD4+CD25+CD127-Treg cells in the blood of tumor-bearing mice(P<0.01).Aerobic exercise could decrease the number of CD4+CD25+CD127-Treg cells in the blood of tumor-bearing mice,but there was no significant difference(P>0.05).(4)Two-way ANOVA analysis of C,OP,E and OPE showed that aerobic exercise combined with OP supplementation had significant interaction with the increase of the expression of microRNA-218 in lung cancer tissue of mice bearing tumors(P<0.05).Simple effect analysis showed that the expression of microRNA-218 in OP group was significantly higher than that in C group(P=0.002,P<0.01).Compared with group E,the expression of microRNA in OPE group was significantly higher(P=0.002,P<0.01).There was no significant difference in the expression of 218(P=0.808,P>0.05);Compared with group C,there was no significant difference in the expression of microRNA-218 in group E(P=0.058,P>0.05).Compared with group OPE,there was no significant difference in the expression of microRNA-218 in group OPE(P=0.335,P>0.05).There was no significant interaction between aerobic exercise and OP supplementation(P>0.05).Aerobic exercise can significantly decrease the expression of miR-21 in lung cancer tissue of tumor-bearing mice(P<0.05).OP supplementation can decrease the expression of miR-21 in lung cancer tissue of tumor-bearing mice,but there was no significant difference(P>0.05).Conclusion:(1)The Lewis lung cancer model of mice duplicated in this study is successful.Aerobic exercise combined with OP supplementation inhibit the growth of lung cancer in mice bearing tumors to a certain extent OP supplementation significantly inhibit the growth of lung cancer in mice bearing tumors.Aerobic exercise also inhibit the growth of lung cancer in mice bearing tumors to some extent.(2)Aerobic exercise combined with OP supplementation mprove the cellular immune function of tumor-bearing mice to a certain extent,and increased the apoptotic rate of lung cancer cells.OP supplementation significantly improve the cellular immune function of tumor-bearing mice and induce the apoptosis of lung cancer cells.Aerobic exercise significantly increase the apoptotic rate of lung cancer cells and inhibitory effect on lung cancer.(3)OP supplementation significantly increase the expression of microRNA-218 in tumor tissues of mice bearing tumors.Aerobic exercise significantly reduce the expression of microRNA-21,thus accelerating the apoptosis of lung cancer cells and effectively inhibiting the growth of tumors.
