Effects Of High-Intensity Interval Training On On Adipose Tissue Metabolomics In Aged Mice

OBJECTIVE:The metabolic and physiologic responses to exercise are increasingly interesting,to investigate the effects of high intensity intermittent training on endogenous metabolites in adipose tissue of aging rats based on GC-MS and to find an effective way of exercise.METHODS:Thirty-six 18-month-old female SD rats were randomly divided into quiet control group(SED),sustained endurance group(MICT)and High Intensity Intermittent Training(HIIT),with 12 rats in each group.In MICT group,treadmill training was performed at a speed of 17 m/min;in HIIT group,treadmill training was performed at a starting speed of 15 m/min for 4 minutes,followed by an increase of treadmill speed to 25 m/min for 1 minute,and nine cycles were performed alternately;in both exercise groups,treadmill time was 45 minutes,five times a week for 8 months;in SED group,rats fed freely but did not train.The body composition of rats was measured by dual energy X-ray 48 hours after the last training.The contents of adiponectin,free fatty acid and inflammatory cytokines IL-6,IL-10,CRP and leptin in serum and adipose tissue were determined by ELISA.Finally,the endogenous metabolites were screened by GC-MS analysis and their changing trends were explored.Result:1)Compared with SED and MICT groups,the lean weight,body fat and the ratio of body fat to lean weight in HIIT group increased significantly;2)Compared with SED group,leptin and serum CRP in adipose tissue decreased significantly in HIIT group(P<0.05),FFA and IL-10 in serum increased significantly(P<0.01),and adiponectin,adipose tissue and serum IL-6 in adipose tissue did not change significantly in each group.3)Analysis of visceral adipose tissue based on GC-MS metabonomics,There were 645 different metabolites in exercise group and SED group.There were 7 significant metabolites in HIIT group and MICT group compared with SED group,including Alpha-Tocopherol,Lumichrome,1-heptadecanol,Palmitic acid,Stearic acid,Octadecadienoic acid and Salicyluric acid.The specific metabolites of MICT were palmitoleic acid and urea,and the specific metabolites of HIIT were oleic acid and cAMP.CONCLUSION:HIIT can significantly improve body composition,inflammation level and adipose tissue metabolism in rats.Palmitic acid,stearic acid and salicylic acid obtained from metabolomic analysis based on GC-MS technology are related to the improvement of inflammation.Metabolites related to oxidative stress include tocopherol,palmitic acid,stearic acid and glycerin.
