Objective: The study of the immunological activity of plant polysaccharides has been a hot topic in the pharmacological research of polysaccharides.The improvement of human mucosal immunity is of great significance for the prevention and treatment of influenza,helicobacter pylori infection,and genitourinary tract HPV virus infection and other diseases.The research object of this article is Radix Saposhnikoviae.Its polysaccharide was seperated and purified,and the the regulation of mucosal immunity of the polysaccharide was studied.Methods: Firstly,three kinds of Saposhnikovia were extracted by classic hot water extraction method and alcohol precipitation method to extract polysaccharides from them,and the differences in polysaccharide content and the distribution of molecular weight were studied.The raw materials that were most suitable for studying the polysaccharide were selected,and the preferred raw materials were then subjected to alcohol precipitation.The alcohol concentration was determined by comparing the polysaccharide content and the distribution of molecular weight.Different concentrations of H2O2 were used for decolorization to optimize the purification process of polysaccharides.Finally,the mucosal immune injury mice as a model,by measuring the concentration of SIg A in different mucosal tissue homogenates,the regulation effect of the polysaccharide on the mucosal immune system was observed.Results: Three kinds of Saposhnikovia materials were compared,and selected Radix Saposhnikoviae for further study.Graded alcohol precipitation and polysaccharide content results showed that 60% alcohol concentration was selected for further study.Decolorization with different concentrations of H2O2 showed that the polysaccharide content was highest after 10% H2O2 decolorization,which was 43.66%.By measuring the concentration of homogenate SIg A in mice with mucosal immune injury model,it is found that the polysaccharide can significantly increase the level of SIg A secretion in mucous membranes of immunocompromised mice.Conclusion:In this paper,we selected Radix Saposhnikoviae as a research medicinal material.This study optimizes the extraction and purification process of polysaccharides from Saposhnikovia,and extracts the polysaccharides from Saposhnikovia.It is found that they have good mucosal immunosuppressive effects and significantly increase the level of SIg A secretion in the mucous membranes of immunocompromised mice.It is suggesting that oral polysaccharides can produce better non-specific effects and increase the ability of mucosal sites to resist microbial infection.It is good for preventing and controlling respiratory,gastrointestinal and urinary tract diseases.
