Food is fundamental for people.It is a substance that maintains normal body functions and continues life.Food safety is related to people's life and health.The successive occurrence of domestic and foreign food safety incidents has made food safety issues a global issue.The issue has been repeatedly banned and has aroused strong concern.Many countries have issued relevant policies.Ingredients in food are extensive and require powerful analytical testing techniques.In today's world,the concept of green chemistry is promoted.Existing analysis methods have problems such as long analysis time,high consumption of organic solvents,poor selectivity,and limited analysis range.Food supercritical fluid chromatography(SFC)has the advantages of fastness,greenness,and good separation performance.In combination with today's popular mass spectrometry methods,it has expanded its application area to a wider range.Potential safety issues exist in both water-soluble synthetic colorants in beverages and biogenic amines in fermented foods,which require attention.Based on the SFC,this article has established methods for the determination of these two types of substances,in order to provide food safety control reference.Chapter ?: The importance and significance of food analysis in daily life are briefly described.The commonly used detection methods and application objects are described.This paper introduces the application progress of SFC in the field of food analysis,analyzes the advantages and disadvantages of current methods,and briefly describes the entry points of this article.Chapter ?: The analysis of water-soluble samples may interfere with the analysis of SFC.In order to detect the doping of artificial colors in beverages and expand the analysis ability of SFC in the field of water-soluble colors,an SFC-DAD method for simultaneous determination of 9 synthetic colorants was established.This method investigated the interaction of water-soluble colorants on six different stationary phases,studied the influence of different kinds of modifiers and additives as well as their respective proportion on the separation,explored their possible effects,systematically selected the outlet pressure and temperature that affect the properties of supercritical fluid,and analyzed the possible causes of the separation effect.The optimum separation conditions of platisil silica(250 × 4.6 mm,5 ?m)column as stationary,methanol(with 0.2% ammonia and 3% water added)as modifier,column temperature of 50 ? and outlet pressure of 100 bar were determined.Under the optimum conditions,the concentration range of colorant was 2.50 ? 450 ?g/mL,the precision RSD was less than 2.0%,the linear correlation coefficient was more than 0.999,and the recovery was between 90.81% ? 100.7%.The optimized method was compared with HPLC and applied to the detection of colorant in carbonated beverage.This method is fast,accurate,selective and environmentally friendly.Chapter ?: In order to detect the content of biogenic amines in fermented foods and avoid the problems of no UV absorption of compounds and complex derivatization steps,an SFC-MS method for the simultaneous determination of nine biogenic amines was established.In this method,the interaction of biogenic amines on eight different stationary phases was investigated.The influence of different kinds of modifiers and additives and their proportion on the separation was investigated.And the influence of different column temperature and outlet pressure on the separation was analyzed.Finally,the mass spectrum make-up solution was investigated.The optimum separation conditions of cosmosil 5HP(250 × 4.6 mm,5 ?m)column as stationary,methanol(with 0.2% ammonia and 5% water added)as modifier,pure methanol as make-up solution,column temperature of 40 ? and outlet pressure of 120 bar were determined.Under the optimum conditions,the concentration range of biogenic amine was 0.10-924 ?g/m L,the linearity of the method is good,the correlation coefficient is more than 0.99,the RSD is less than 4.50%,the precision is less than 9.36%,and the recovery is between 75.82% and 99.93%.The optimized method can be successfully applied to the determination of biogenic amines in 10 brands of soy sauce samples.This method is fast,accurate and less organic solvent consumption.It successfully overcomes the problems of no UV absorption and complex derivatization steps.Chapter ?: The content of the work done is summarized,and the improvement direction is prospected according to the deficiencies in the work.
