Food is essential for one to keep healthy and alive,and the quality of food is related to one's health.Therefore,it's of great significance to make sure that the publicly sold food was high-quality products and harmless.However,in recent years,there have been many such food quality and safety incidents.Food analysis is performanced to ensure food quality but it's difficult to be conducted sometimes since food samples are complex and contain diverse ingredients,making its analysis and detection a certain degree of challenge.Various analytical methods,such as high preformance liquid chromatography(HPLC),gas chromatography(GC),etc.,have been developed for food analysis.However,these methods have some disadvantages,such as long analysis time,large consumption of organic solvent(HPLC)and required derivatization process(GC).Therefore,it's meaningful to develop new analytical method.Supercritical fluid chromatography(SFC)has been widely employed in food analysis,for the determination of food components such as lipids,adulterants,phenols and carbohydrates,due to its advantages such as fast separation,lower solvent toxicity and better environmental impact.However,the applications mainly focus on chiral separation and analysis of low polarity of compounds,quite few studies have been reported about the analysis of polar compounds by SFC.Therefore,in this dissertation,rapid,sensitive and efficient SFC-MS methods have been developed to determine polar compound in food,such as phenolic acids,saponins,biogenic amines etc.Chapter ?: The importance of food quality and food analysis were introduced systematically,and existing analytical methods are reviewed.What's more,the features of SFC and its applications in food analysis are highlighted,and the research strategy and innovations of this dissertation are described.Chapter ?: a fast,sensitive and highly efficient SFC-MS method was developed for the determination of 12 phenolic acids in EVOOs.The separation conditions including the stationary phase,the mobile phase additives,the composition of the make-up solution,the temperature and back-pressure,were systemically optimized.The SFC method was also compared with an HPLC method in terms of analysis time and selectivity.The developed SFC-MS method was validated and applied to the analysis of phenolic acids in EVOO extracts.Chapter ?: An efficient and rapid SFC-MS method was established to simultaneously analyze eleven polar compounds(including 4 flavonoids,3 saponins,and 4 organic acids)in Lonicerae japonicae flos and Lonicerae flos.All the factors affecting the chromatographic separation,such as the type of the stationary phase,the type and concentration of the additive,and backpressure were systematically optimized.The results of real sample analysis showed that the developed method could be applied to the identification of Lonicerae japonicate flos and Lonicerae flos.Chapter ?: Since the underivatized biogenic amines are strong basic compounds and difficult to be analyzed by HPLC and other methods,a SFC-MS method was developed in this chapter.Under optimal separation conditions,effective separation of nine biogenic amines was achieved within 25 minutes.The peak tailing of biogenic amines existing in previous methods was overcame and the Cumbersome,time-consuming derivation process was avoided.The developed method was applied to the analysis of biogenic amines in eggs.Chapter ?: The research work of this dissertation was summarized and the future outlooks of this research were also discussed.
