African violet,Gesneriaceae genus of African violets,Latin name:(Saintpantia ionantha Wendl),also known as the African violet or saint including flowers,perennial evergreen herbs,color is very rich and loved by the people.In this experiment,the leaves of 2 species of African violet were used as materials,and the regeneration system of African violet was established and optimized.The optimum formula of bud proliferation and rooting medium was obtained.The best callus induction medium was MS+0.5mg/L6-BA+0.4mg/LNAA+0.2mg/L2,4-D;the best differentiation medium was MS+0.5mg/L 6-BA +0.1mg/L NAA;the best rooting medium was 1/2MS+0.5 mg/L IBA +0.5mg/L NAA.In the experiment of colchicine in vitro induction of African violet leaves,in order to find the most suitable and the African violet colchicine concentration and time were determined by different concentration of regenerated plant physiological and biochemical indexes of African violets,later got strong resistance,high ornamental value and excellent new varieties of resources for African violets base.The test has made some progress,the experimental results are as follows:(1)to test medium with 10mg/l concentration of colchicine treatment for 20 days for the best performance,compared with the control group,after colchicine treated buds,partial leaf hypertrophy,dark green leaves,the plant is large,another part in flower and leaf buds;(2)in the20 d treatment,the content of soluble sugar and soluble protein were higher than CK,that after colchicine treatment can improve the metabolic capacity;MDA and SOD in 20 d,10d and 30 d resistance is good,can effectively remove oxygen free radicals within the plant tissue,improve the environmental defense ability have stronger,cold and drought resistance,disease resistance.
