Background/Purpose:Prostate cancer(PCa)is one of the most common malignant tumors in male worldwide,particularly in developed western countries.Long non-coding RNAs(IncRNAs)have emerged as important regulators and biomarkers of tumor development and progression.This study aimed to investigate the clinical significance,biological functions and the underlying mechanism of lncRNA-LINC01296 in prostate cancer.Materials and Methods:IncRNA-LINC01296 expression was detected by quantitative real-time PCR(qRT-PCR)in prostate cancer tissues and cell lines.Its association with clinicopathological characteristic of prostate cancer was analyzed using the Kaplan-Meier analysis and the Cox proportional hazards model.Small interfering RNA was used to suppress lncRNA-LINC01296 and the knockdown efficiency was examined by qRT-PCR.Cell Counting Kit-8 assay,colony formation assay,migration and invasion assay and western blot were used to further explore its role in tumor progression.Results:The expression level of IncRNA-LINCO 1296 was higher in prostate cancer tissues and prostate cancer cells compared to adjacent non-tumor tissues and normal prostate stromal immortalized cells WPMY-1.Its expression was correlated with Preoperative PSA(P=0.002),Lymph node metastasis(P=0.035),gleason score(P=0.000),Tumor stage(P=0.036).The patients with higher IncRNA-LINC01296 expression had an advanced clinical features and a shorter biochemical recurrence-free survival time.And,multivariate analysis showed that the status of IncRNA-LINC01296 expression was an independent predictor of biochemical recurrence-free survival in prostate cancer.Additionally,knockdown expression of IncRNA-LINCO 1296 could inhibit PCa cells proliferation,migration,and invasion in vitro study through regulating PI3K/AKT/mTOR signalling and EMT process.Conclusions:Our results demonstrated that the expression level of IncRNA-LINCO 1296 is closely related to the biochemical recurrence of PCa patients.Increased level of IncRNA-LINC01296 promotes prostate cancer growth and metastasis by activating PI3K/Akt/mTOR signaling pathway and EMT process.
